The Anti-tumor Effect And Its Mechanism Of Huajiyi-ganjian And 5-fluorouracil Combination To H22 Tumor-bearing Mice

Objective: This paper mainly research on the anti-tumor effect to hepatocar- cinoma22 tumor-bearing mice in the Chinese traditional medicine huajiyiganjian,Che- motherapy drug 5-fluorouracil and the combination effect of huajiyiganjian and 5- fluorouracil; preliminary discuss the express influence of huajiyiganjian to Proteins such as TGF-a,EGFR,Caspase-3 .Methods: The liver cancer model has been set up and stochastically divided to four groups, which includes normal saline stomach perfusion,Huajiyiganjian stomach perfusion,5-fluorouracil intraperitoneal injection and the combination effect of Huajiyi- ganjian stomach perfusion and 5-fluorouracil intraperitoneal injection. The mice were conventional feed and sacrificed after 14 days. It's tumor tissue,spleen and thymus gland were peeled off and further weighed after the filter paper drying process. The tumor growth inhibition rate spleen index and thymus gland index were deter- mined separately. after the processes of 10% Formaldehyde fixation, dehydration,transpar- ence,waxing,embedment and paraffin section, the tumor tissue were carried out by the immune dyeing experiment to determine the protein express of TGF-a,EGFR,Caspase- 3 in the tumor tissue.Conclusion: (1): Treatment group: tumor inhibition rate of Huajiyiganjian,5- fluorouracil and the combination one are 44.23%,35.74% and 54.83%。Compare with the model groups (normal saline), treatment group has remarkable difference (P<0.05 or P<0.01). (2): The combination of Huajiyiganjian and 5-fluorouracil will increase the tumor inhibition rate in some extent, which reach to 54.83%.(3): Treatment group can reduce the express of TGF-a and EGFR protein in tumor issues, in which, the com- bination group has more treatment effect. (4): Treatment group can add the express of Caspase-3 protein in tumor issues, in which, the combination group has more treatment effect. (5): The influence of Huajiyiganjian to spleen index and thymus gland index of tumor-bearing mice: compare with 5-fluorouracil, Huajiyiganjian has a remarkable improvement(P<0.05).Results: Huajiyiganjian, which has the function of heat-clearing and detoxifying,softening and resolving hard masses,strengthening spleen and replenishing qi,support foundation and clear virus, is a experienced prescription of primary liver cancer treat- ment created by the writer's advisor for five years clinic experience. This experiment preliminary confirm that Huajiyiganjian and its combination with chemotherapeutic drugs can control the growth of tumor in some extent and remarkable increase the tumor inhibition rate than chemotherapeutic drugs only. Its anti-tumor mechanism may related to the control of TGF-a,EGFR express and increase of Caspase-3 express in tumor-bearing mice; Huajiyiganjian can protect the spleen and thymus gland of tumor- bearing mice, its mechanism will probably be that this medical can remarkable control the immune organs atrophy, which resulted by the chemotherapeutic drugs, on the other hand, this medical may also has the ability to increase the immune function of tumor-bearing mice. In all, the anti-tumor mechanism of Huajiyiganjian will be re- searched further.
