Objective: To study the mechanisms of acupuncture analgesic (AA) induced by musice electroacupuncture(EA)and production of electroacupuncture tolerance. Methods:The rats were randomly divided into control group,pulse EA group,music EA group.(1)The tail flick latency(TFL) caused by hot water was used as pain threshold.The percentage increased in pain threshold was taken to assess the efficacy of EA analgesia. (2)Electrophysiology:Trains of the electric impulses were applied to the sciatic nerve. The discharges of pain-related neurons of CPU in rats were recorded with glass microelectrode extracellular. (3)Immunohistochemistry: imrnunohistochemical method was performed to measure the change of Cholecystokinin octapeptide(CCK-8) positive expression in CPU in rat. Results: (l)After EA 1-6 hoursmusic electroacupuncture could prolonged TFL and .Music EA not only inhibited the discharges of PEN,but also excited the discharges of PIN. (2)Pulse EA possessed AA afert EA 1-3 huors,but after four hours,TFL and the discharges of pain-related neurons recoverd the degree of preEA. (3) After EA 1-6 hours Music EA reduce CCK-8 positive expression in CPU,but pulse EA enhanced it. Conclusion: The above results suggested that music EA can raise the pain threshold and keep AA for a long time.
