| INTRODUCTION Asthma is a commonly encountered disease in respiratory system ,the incidence of asthma worldwide has increased significantly over the past two decades.For a long time ,exploring the pathogenesis of asthma had been limited the degrulation of mast cells caused by allergen.Recently ,it has been showed that airway inflammation plays a pivotal role and that airway hyperresponsiveness has involved in the pathegenosis of asthma .Therefore ,asthma is a chronical airway inflammation characterized by the infiltration of inflammatory cells including mast cells,eosinophils and T lymphocytes and so on,accompanized by airway hyperresponsiveness. Traditional Chinese Medicine(TCM) calls asthma whezzing syndrome.During the relax phase of asthma ,lung deficiency,renal asthenia and spleen deficiency are a principal cause of asthma .Spleen is the source of phlegm.Over-fatigue and improper diet leads to spleen deficiency which causes failure of spleen in transportation of fluid .With retention of fluid and cold intermingled with heat,turbid phlegm is produced.On the other hand,spleen deficiency leads to insufficiency of pulmonary qi which is responsible for defensive exterior to exogenous pathogenic factors' invade.As a result of spleen deficiency andinsufficiency of lung qi ,asthma is induced.It has been documented the effects of spleen deficiency syndrome on asthma as followsrincrease total counts of WBC and eosinophils counts in BALF of guinea pig with asthma;aggravate the imblance between oxidation and anti-oxidation of asthma;descent cAMP/cGMP ratio value in BALF of rats with asthma.However ,to date ,the effect of spleen deficiency syndrome on airway hyperresponsiveness of asthma has not been studied.To date ,Glucocorticoids(Gcs) has been the most effective drug in asthma therapy ,but the treatment of Gcs for long time and high dose has been associated with serious side effect at the same time. People try to replace the Gcs with other anti-inflammatory drugs. According to systemic modilation and treatment advantage of Traditional Chinese Medicine (TCM) for asthma therapy, the development of drugs from Chinese medicine herbs used for asthma has been of important significance. Cryptoporous volvatus is the sporophore of Cryptoporous volvatus [(Peck) Schear] which has been used as the folk remedy for tracheitis and asthma because of its activities of expelling wind and eliminating dampness, and dredging meridians to activate blood. Recently, the effect of polysaccharides of ferment cultured Cryptoporous volvatus on inhibiting release of leuketriences from lung and anti-allergy in sensitized/challenged guinea-pig has been documented.another study has demonstrated that Cryptoporous volvatus ferment substance (CVFS) can inhibit eosinophilic chemotaxis induced by PAF. It has been showed that CVFS can be used for asthma therapy because of its anti-inflammatory and anti-allergen effects. However ,the experimental study on the effect of CVFS on asthma systemically has hardly been documented.AIM The objectives of present study were as follows: (1) the effect of spleen-deficiency syndrome on the allergen-induced asthma. (2) whether the effect of CVFS on allergen-induced asthma in mice with asthenia of spleen .(3) primary study on mechanism of CVFS. METHOD NIH mice weighing 20±2g were used. They were sensitized by asingle im of ovalbumin 0.1 mg mixed with alumin hydroxide 10 mg in saline 0.1 ml at two back footpads on 1d.They were administrated with CVFS (ig) 0.3,0.9,2.7g.kg-1 on 9d,and one time every day for 13d. However ,positive-drug group were administrated oral Radix Astragali recipe 0.5ml per mouse,and model group with saline 0.5ml per mouse at the same time .They were sensitized intraperitoneally by the same antigen on 11d.Mice with asthenia of spleen was reproduced on 12d.The treament is as follows: the mice were feed with pig fat 30ml.kg-1 and swam in warm pool until over fatigue after feed ,one time everyday for 10d.Mice were challenged by expoure to an aerosol of ovalbumin 10g.L...
