ObjectiveThe pharmacodynamics of Kadsura heteroclita on hippocampal neurons and the Alzheimer's disease (AD) model mice were studied. Methods(1)The rat hippocampal neurons were cultured with 0.2,1.0mg · mL-1 HS2.The convert phase microscope was used to analyse the number and the length of neuritis, as well as the maximum diameter of hippocampal neurons soma. The survival neurons count and lactate dehydrogenase (LDH) assay were carried out in different culture time. (2)The experiment of HS2 maximum tolerance dose is made for mouse (po). (3) The Kung Ming mice were treated with D-galactose and AlCl3 (90d) to make the AD animal model. The therapy groups were treated with HS2 (po, 40d) respectively since d51. The Morris water-maze test, analysis of Ach and AchE activities, the expression of APP, PS1, BACE, as well as the pathological observation of the hippocampus and cerebral cortex were carried out to evaluate the effect of HS2 on the AD model mice. Results(1)1.0mg · mL-1HS2 could promote the growth of neurons. (2)The MTD of HS2 is 126 g · kg-1.(3)By oral administration with HS2, the capacity of learning and memory of the AD model mice was improved; the content of Ach and the activities of AchE reached the normal levels. HS2 can downregulate the expression of APP, PS1, and BACE and decrease the amount of senile plaque in their brains. ConclusionsHS2, 1.0mg · mL-1 , can promote the growth of hippocampal neurons. HS2 can meliorate AD model mice' learning and memory deficiency and sustain the normal levels of Ach, AchE, as well as decrease the mRNA content of APP, PS1, BACE and senile plaque in their brains.
