| Purpose: Leptin, interleukir-6 (IL-6) , Ca2+, P", B-ALP and BGP was detected. On the same time using DEXA to detect BMD. Then try to find out the relationship between the leptin, IL-6, Ca2+, P", ALP, BGP and osteoporosis.Method: Choosing 120 female SD rats in 6 months old, randomly divided them into 6groups (obesity control group, obesity vacuity group, obesity castrated group, normal body weight matched control group, normal body weight vacuity group and normal body weight castrated group). There are 20 rats in each group. The detection process will proceed from the 2nd, 3rd, 4th, 5th and 6th month respectively. All the groups belonging to the normal body weight were fed in basic feed and all the obesity groups were fed in hyper-fat feed. The feed was referred to "The check-out method of the evaluation method of the healthy food functiology"( published by the health supervision of the ministry of health). Every 2 rats were fed in one cage, free for drinking water, but limited ingestion, that can make sure the appetites of all the groups have the same quantity of feed. The room temperature was kept in 25 ? 癈.Every week to measure their weight and count the average weight. The experiment group (castrated group) was anesthesia infra-abdominal cavity by using 10% chloral hydrate (3ml/100g) and proceeded the bilateral ovaries transfer from the abdomen. All the control groups were taken the ovaries out and then put the ovaries back to the abdomen again. All the vacuity groups were doing nothing. All the rats were fed as before after sugary. Took out 4 rats after the surgery in the 2nd, 3rd, 4th, 5th and 6th month randomly and detected the bone mineral density by useing the Dual X-ray absorptiometry, withdrew blood from the heart, then got the blood serum by using the centrifugation and detected the leptin and IL-6. Using the biochemistry assay to detect the level of Ca2+, P" and ALP. Using the radioimmunoassy to detect the level of BGP. All the data were recorded by SPSS 10.0 and then proceed bi-agent analysis of variance by using the statistics software (random area group designed) .Result: The order of leptin from high level to low level is that obesity castrated group > obesity vacuity group > obesity control group > normal body weight matched control group >normal body weight castrated group > normal body weight vacuity group. The order of BMD from low level to high level is that obesity castrated group < normal body weight castrated group < obesity control group < obesity vacuity group < normal body weight vacuity group < normal body weight matched control group. The order of Ca2+ from high level to low level is that obesity castrated group > obesity vacuity group > obesity control group > normal body weight castrated group > normal body weight matched control group > normal body weight vacuity group. The order of P" from high level to low level is that obesity castrated group > normal body weight castrated group > obesity vacuity group > obesity control group > normal body weight vacuity group > normal body weight matched control group. The order of B-ALP from high level to low level is that obesity castrated group > normal body weight castrated group > obesity control group > obesity vacuity group > normal body weight vacuity group > normal body weight matched control group. The order of BGP from high level to low level is that obesity castrated group > normal body weight castrated group > obesity vacuity group > obesity control group > normal body weight matched control group > normal body weight vacuity group. The order of IL-6 from high level to low level is that obesity castrated group > normal body weight castrated group > obesity control group > obesity vacuity group > normal body weight matched control group > normal body weight vacuity group. The result cue that leptin can caused or increased osteoporosis.Conclusion : There is obverse relationship between the osteoporosis and theconcentration of the leptin.
