Objective: We report that the effect of basic fibroblast growth factor (bFGF) on the dentate gyrus (GD) and CA3 region of hippocampal formation from the pilocarpine-induced model of chronic seizures.Methods: Wistar rats was divided into two control groups and four experimental groups( I , II ,III, IV ). All experimental rats were injected pilocarpine; and these of the rats injected with bFGF in ventriculus dexter cerebri, and then injected with pilocarpine in intraperitoneal after 2 hours were given bFGF; Animals were allowed to survive for 15 days in experimental groups III , IV. All rats anaesthetized and processed for brain sections that were taken through the hippocampal site completely, the effects of experimental were obsereved or determinined in both by H-E staining (with light microscoply) and Immunohistochemical staining for glial fibrillary acidic protein(GFAP), unit area of astrocytes were determinined in four experimental groups for hippocampal neuropil with optical mean of gray.Results: The respiration swiftness began to appear at 5-7min after pilocarpine injection, and respiration rate is counted at a mean of 81.03 ?5.94once.min-1. all rats were observed for occurrence of behavioral change that evolved to repeated generalized clonic seizures during the time(similar to kindle stage 5 seizures in with Racine) . Of the animals injected with pilocarpine, the kindle time at a mean of 25.36?.24(min) in experimental group I and II which 64.3% developed II-V stage , and at a mean of 19.52 + 5.59 (min) in experimental group III, IVwhich 84.2% developed II - V stage and then which no clear behavioral3seizures were observed in latter 2-15 days after first seizure 0Examination of HE-stained with experimental groups I in hippocampal revealed a widespread damage(lead to cell necrosis, cell degeneration) including the dentate gyrus (GD) and CAS region. The experimental group III in the hilus of the dentate gyrus (DG) were asignificant reduced and especially in. There was a cells degeneration in experimental group IV, but which shown regular on the distrabuiion.The result of experimental groups from the astrocyte and GFAP-po-sitive cells of hippocampus with the immunohistochemistry method shown that there was significant difference for average area of astroglia(A), Vv, VvcB and Nv. Experimental group I in CAS region compare with other experimental groups, Vv, P<0.05; A , VvcB and Nv, P<0.01. and in GD, P< 0.001, The average area of astroglia(A) of GFAP-positive cells in the granule cell layer of the DG , groupsllland IV ,groups II and IV, P<0.05. A highly significant correlation was noted between the CA3region and the granule cell layer of the DG for four groups. The Vv of GFAP-positive cells of hippocampus by analysis of variance showns that GD in groups I , III, P<0.01; and CA3 region in groups I , IV, P<0.01. The data indicate that the time-related of the hippocampal neurons in seizure respond differentially to bFGF.Conclusion: These results demonstrate that bFGF is useful factor for the neuronal survival and neurite extension that occur in epileptic model by pilocarpine-induced one. It was also suggested that bFGF can produces a significant increase in number of neurite-bearing cells, as well as in the degree of their fiber network . Moreover, It is showed that even though a certain amount of cell loss in specific areas may be essential for chronic seizures to occur, excessive cell loss may hinder epileptogenesis, such as found SE.
