The objective of this investigation was to compare the pharmacodynamics of Sheng Wu Niu Huang (SWNH) with those of the natural calculus bovis and the artificial calculus bovis from the acute toxity, antipyretic effect, hypnotic effect, anticonvulsive effect, anti-inflammatory effect, antiviral effect and the influence on the hemorheology. The results showed that the acute i.g. LD50 of SWNH in mice was more than or equal to 10,000 mg/kg with the natural calculus bovis and the artificial calculus bovis as lower toxic drugs. The acute i.p. LD50 of SWNH made by 29#and 30# bacteria were 244.91 mg/kg and 278.04 mg/kg respectively, being different from that of the natural calculus bovis (675.77 152.05 mg/kg), and that of the artificial calculus bovis (more than or equal to 1,000 mg/kg). SWNH (25, 50, and 100 mg/kg) and the artificial calculus bovis (50 mg/kg) i.g. lowed the body temperature significantly compared with the control group in the fever model induced by 2,4-dinitrophenolate, while the natural calculus bovis(50mg/kg) did not. SWNH (100, 150 mg/kg) i.p. prolonged markedly the time of sleep in mice with the hypnotic effect induced by phenobarbital sodium, while SWNH (50 mg/kg), the natural calculus bovis (100 mg/kg), and the artificial calculus bovis (100 mg/kg) did not. SWNH (500, 1,000, 2,000 mg/kg), the natural calculus bovis (1,000 mg/kg), and the artificial calculus bovis (1,000 mg/kg) i.g. did not prolong significantly the latency of Annaka-induced convulsion in mice, with no marked difference among these groups. SWNH (50,100,150 mg/kg), the natural calculus bovis (100 mg/kg) i.p. had similar effects on the xylene-induced inflammatory in mice. The effects of these groups were smaller than those of the artificial calculus bovis (100 mg/kg), and the control group, but not significantly. The drug-containing serums made by SWNH (25,50,100 mg/kg), and the artificial calculus bovis (50 mg/kg) i.g. in rats respectively, after being inactivated and diluted 10 times, increased the TCID50 of New castle I strain vaccine virus on the chicken embryo fibroblast(CEF) by 1 titer, compared with the negative control group. While the drug-containing serums of the control group and the natural calculus bovis (50mg/kg), with the same treatment, decreased the TCID50by 1.5 titers. SWNH (25,50,100 mg/kg) i.g. decreased significantly the erythrocyte sedimentation rate (ESR), K value of erythrocyte sedimentation equation, and the red blood cell (RBC) aggregation index (Al) in rabbit models of blood stasis syndrome induced by Dextran. While the natural calculus bovis (50 mg/kg) decreased significantly the ESR and K value of erythrocyte sedimentation equation, and the artificial calculus bovis (50 mg/kg) decreased significantly the ESR.
