ObjectiveTo observe the effects of natural calculus bovis and calculus bovis cultivated by glucuronidase on antiinflammatory, anticonvulsion,analgesia, sedative,hypnosis and their protective effect on anti-hypoxia in mice.To estim- ate the pharmaceutical value of calculus bovis cultivated by glucuronidase and provide evidence for finding the substitute for natural calculus bovis.Methods1.Sedative and hypnosis experimentTo observe the influence of Natural Calculus Bovis(NCB) and Calculus Bovis Cultivated by Glucuronidase(CBCG) on active motion in mice by method of photoelectricity calculation. Adopting pentobarbital sodium (threshold dose)-induced hypnosis experiment to observe the effect of NCB and CBCG on hypnosis in mice. 2.Anticonvulsion experimentApplying Nikethamide(NTM)-induced convulsion model and maximal electroshock convulsion(MES) model to observe the effect of NCB and CBCG on anticonvulsion in mice.the experiment shows that both NCB and CBCG can inhibit convulsion in mice.3.Antiinflammation experimentTo adopt CGN-induced foot inflammation model in mice and Dimethyl- benzene (DMB)-inducde inflammation model in mice to observe the effect of natural ca lculus bovis and calculus bovis cultivated by glucuronidase on inflammaation.4.Analgesia experimentAdopting acetic acid-induced writhing experiment in mice to observe the effect of natural calculus bovis and calculus bovis cultivated by glucuronidase on writhing times of mice.To adopt recording instrument of mice active motion to measure the amount of activity of mice after administrating medicine.To observe the effect of natural calculus bovis and calculus bovis cultivated by glucuronidase on active motion of mice. Applying light radiation- induced tail flick in mice experiment to survey the effect of natural calculus bovis and calculus bovis cultivated by glucuronidase on threshold value of pain of mice.5. Anti-hypoxia experimentAdopting normal and ISO-induced hypoxia in mice experiment to observe the effect of natural calculus bovis and calculus bovis cultivated by glucuro- nidase on anti-hypoxia in mice.Results1. The hypnotizing experiment indicates that both natural calculus bovis and calculus bovis cultivated by glucuronidase can pr olong the sleep time of mice.Meanwhile,the two can strenthen the hypnosis effect of continal in mice.2. the outcome of anticonvulsant experiment shows that calculus bovis cultivated by glucuronidase can relieve convulsion.3. The results of antiinflimmatory experiments show that both naturral calculus bovis and calculus bovis cultivated by glucuronidase can inhibit DMB-induced ear inflimmation and CGN-induced foot inflimmation in mice.But the two have different inhibiting rate of inflimmation.4. The result of analgesia experiment shows that both natural calculus bovis and calculus bovis cultivated by glucuronidase can reduce writhing frequency in mice.Both can raise pain threshold value of mice.5. The experiment shows that calculus bovis cultivated by glucuronidase can strenthen the ability of anti-hypoxia of mice.Conclusions1.Both NCB and BCG possess the effect of central nerve system suchl as antconvulsion,sedative,hypnosis,etc.They can refrain convulsion from happe- ning in mice.2.NCB and CBCG possess the effect of antiinflammation,analgesia. They can repress DMB-induced inflammatiom in mice and relief the grief induced by CA and light-radiation in mice.3.CBCG can enhance the ability of antihypoxia in mice and prolong the survival time of mice. 4.The above results show that CBCG possesses the effect of natural nerve system as good as NCB. CBG may replace NCB on the pharmaceutical effect in natural nerve system partly.
