ObjectiveStudy the effect of astragalus polysaccharides ( PG2) on 6 kinds of antiphospholipid antibodies { anticardiolipin antibody, antiphos-phatidyl serine antibody, antiphosphatidyl inositol antibody, antiphos-phatidyl ethanolamine antibody, antiphosphatidic acid antibody and antiphosphatidyl choline antibody) , urine protein and survival time in NZB x NZW Fl mice, investigate the different function of different dosage of astragalus polysaccharides.Methods1. 24 NZB x NZW Fl female mice were randomly divided into 4 groups; 7 in group PG2I (25mg/kg) , 6 in group PG2II (50mg/kg) , 5 in group dexamethasone ( Img/kg) , 6 in group NS (0. 2ml/d) , which were injected once a day. Select 16 lupus - prone mice BXSB and 12 normal C57BL/6 mice as contrast.2. Antiphospholipid antibodies were detected by ELISA in the serum of these mice.3. Bradford method was used to detect urine protein.4. When they were dead, the survival time were recorded.5. Statistical analysis; t test was used to analysis the difference of antiphospholipid antibodies and urine protein among these groups. Log - rank test was used to analysis survival time.ResultsI. Antiphospholipid antibodies1. The optical density of antiphospholipid antibodies in group NS were higher than that of BXSB and C57BL/6 mice ( P <0. 05 or P < 0.01).2. Compared with group NS, the optical density of antiphospholipid antibodies in group DEX decreased significantly ( P < 0. 05 ) ; There was no difference of OD value among group DEX, BXSB and C57BL/6.3. The optical density of antiphospholipid antibodies in group PG2I increased significantly compared with BXSB and C57BL/6 mice (P<0.05 orP<0.01).4. Compared with group NS, the optical density of antiphospholipid antibodies in group PG2II decreased significantly ( P < 0. 05 ) . There was no difference of OD value among group PG2II, DEX, BXSB and C57BL/6.II. Urine proteinThe urine protein of group DEX, PG21, and PG2II were significantly decreased compared with that of group NS.M. Survival timeThe mice in group NS have all been dead when they are 15 months old. There are 1 in group DEX, 2 in group PG2I and lin group PG2II still alive until 20 months. The survival time of these groups hasno significant difference ( P = 0. 1248 ).Conclusion1. Except for aPI, 5 kinds of antiphospholipid antibodies in NZB x NZW Fl mice were significantly higher than that in group BXSBand C57BL/6.2. The optical density of antiphospholipid antibodies of SLE mice in group DEX decreased significantly compared with that of group NS. We can draw a conclusion that as an immunosuppressive drug, dexam-ethasone can inhibit the generation of antiphospholipid antibodies.3. Compared with group NS, antiphospholipid antibodies increased in group PG2I and decreased in group PG2II. This indicated that low - dose of PG2 can promote the generation of antibody, and high - dose of PG2 can act as an immunosuppressive drug.4. Dexamethasone and astragalus polysaccharides can decrease urine protein and improve the injury of kidney of SLE mice.
