PrefaceAtherosclerosis (AS) is a kind of disease which do harm to the health of human beings severely . Its incidence is 79.9% in Chinese people whose ages are more than 60. Arterosclerotic lesions usually develop at great or mid type arteries. Atherosclerosis obliterans( ASO) was found at iliac , femoral and popliteal artery especially. With the improvement of living conditions and the development of diagnosis technology of vascular surgery, the current of ASO is increasing and the age of it is going to be younger gradually . But the pathogery of ASO has not been clear completely. According to epidemiology investigation , the risk factors were considered as hyperglyceridemia, hyper-tention, smoking, diabetes smellitus and inheritation. Recently some scholars advanced that infection might impact the development or progression of AS, and chlamydia pneumoniae (Chp) might has relation to arteriosclerotic lesions. In this study the DNA of Chp was detected by polymerase chain reaction (PCR) in order to investigate the relationships between the expression levels of Chp DNA and ASO in low limbs. It will play a positive role in the treatment of ASO.MethodsThe venous whole blood (2ml) was collected in EDTA -treated tubes from 60 patients who were admitted to the vascular department of the second hospital affiliated Shenyang Medical College because of ASO, all the blood samples were stored at -20c. The control group comprised 40 healthy persons. The DNA of Chp in venous blood was detected by PCR analysis. The extrated DNA was used in PCR round, and the products were measured by polyacrylamide gel electrophore-sis. Analyze the electrophoretic images by statistics, P<0.05 was regarded as significant.ResultsThe positive expression rate of Chp DNA was 78. 33% (47/60) in 60 cases with ASO, while the positive rate was 47.50% (19/40) in 40 cases as control group( P <0.05). In all 60 cases , there were rises in the positive rates of Chp DNA from I stage to IV stage, the positive expression rate of ChpDNAwere33. 33% ,73.68% ,86.96% and 91.67% respectively, the difference between I to II stage and III to IV stage was significant (P<0.05).DiscussionsASO is a kind of multifactorial disease. The risk factors conclude hyperglyceridemia, hypertention, smoking, diabetes smellitus and in-heritation. In the resent years some experts have considered Chp as a factor of arterosclerotic lesions.1. The effect of Chp on the occurrence of ASOThe mechanism that Chp causes AS may be the following explain-ations: Chp enters into blood from respiratory tract and is transmitted to blood vessels. It can infect phagocyte, pavement corpuscles and smooth muscles, then proliferate in them. The polysaccaride of Chp combines with low density lipoprotein( LDL). So the antibody or toxin is produced, this induces AS. In this study the expressive level in patients with ASO was higher than that of the control group, so there might be certain relationship between Chp and ASO.2. The relationship between Chp and the serious extent of ASO In all 60 cases with ASO the positive rate of Chp rised with thedevelopment of ASO . It shows that Chp has relation to the extent of ASO. The expression of Chp DNA depends on the lesion of artery, the distal artery of obliteration and the extent of lesion in laterad artery.3. The clinical significance of the Chp detectionThe macrolide antibiotic is a kind of drug that is used to treat Chp infection. This study demonstrated the relationship between Chp DNA and ASO. It provided the evidence to treat ASO by macrolide antibiotic. And in this study it was clear from genetic field.Conclusions1. The positive expression rate of Chp DNA was higher than that of the control group. It suggested there were some relationships between Chp and ASO in low limbs.2. The positive expression rate of Chp DNA influenced the development of ASO.
