It is a difficult problem that denervated skeletal muscle atrophy prevents limbs function from recovering. Author regards that the cause of unreverted skeletal muscle atrophy probably is increment of connec-tive tissue. The experiment use bFGF compound agents to delay the speed of skeletal muscle atrophy and restrain increment of connective tissue at the same time.Material and method1. Material56 mouse are divided into 4 groups , their left limb is experimen-tal side and right limb is contract side. Silicon glue tube with bFGF and connective fiber restrain agents are put into denervated gastrocne-mius separately in A group. 2 Silicon glue tubes with connective fiber restrain agents are put into denervated gastrocnemius in B group. 2 empty Silicon glue tubes are put into denervated gastrocnemius in C group. Nothing is put into denervated gastrocnemius in D group.2. Operation methodAt first stomach anaesthetize is conducted, then sciatic nerve is found and is cut break in the same site. At last Silicon glue tube is put between muscle and muscle member.Observation index4 groups gastrocnemius are treated and Observed in 30 and 90 days after operation.1. Electromyography inspection : Observe the change of fibrilla-tion potential amplitude in 4 groups gastrocnemius.2. Gastrocnemius weight measurement3. Histology section observation: H. E, Mallory , Vitamin C sliv-ery stain, SDH4. Electric microscope inspection5. Immuno - histology section of myodynamic protein6. Microimage analyzer observationResults1. Results of observation with naked eyesRemarkable atrophy change can be observed in denervated skele-tal muscle group. The comparative results of each group: ( 1) The size of A, B group gastrocnemius is larger than that of C, D group. The degree of A, B group gastrocnemius adherence is slight than that of C, D group. (2) The size of A group gastrocnemius is larger than that of B group. The degree of A, B group gastrocnemius adherence is slight than that of B group. There is no remarkable difference between C and D group. These are the evidences of the function of the agents this experiment used, the function is prolonging muscle atrophy and preventing increment of connective tissue.2. Observation with microscope (1) MicroscopeRemarkable atrophy change can be observed in denervated skele-tal muscle group in H. E, Mallory and Vitamin C slivery stain section: the muscle fibers of A,B group are more complete , thicker, larger number , with more nuclei and clearer cross - lines than C, D group. The degree of A, B group gastrocnemius adherence is slight than that of B group. There is no remarkable difference between C and Dgroup. Things are the same according to the ratio of white and red muscle fibers in SDH section.(2) Electric microscope inspectionThe degeneration changes can observed in each denervated skele-tal muscle group , such as muscle silk disorder , chondriosome disap-pear , hepatin reduction. But we find more these kinds of degeneration in C, Dgroups than in A, Bgroups.3. Gastrocnemius weight measurementAverage muscle weight of A,B group is more than that of C,D group and the difference is meaningful in statistics.4. Microimage analyzer observationThe agents this experiment used have the function of prolonging muscle atrophy and preventing increment of connective tissue.5. Electromyography inspectionThe fibrillation potential amplitude of A, B group is larger than that of C, D group, there is no remarkable difference between C and D group.Discussion1. bFGF compound agents has the function of preventing dener-vated skeletal muscle atrophyAuthor regards that the cause of unreverted skeletal muscle atro-phy probably is not mainly the nerve control lose but is the increment of connective tissue.
