ObjectiveTo prepare tissue engineered bone graft loading titanium dental impant.Methodstitanium dental impant ( 3 mm in diameter) was inserted into cancellous bone matrix ( CBM ) column ( 5 mm in diameter). Bone marrow derived osteoblasts were cultured and expanded in vitro. Cells were induced by recombinant human bone morpnogenetic protein - 2 for thtee days and then harvested and seeded into porous CBM and on-to dental impant at the density of 2 l08Vml. Four cell - coral - impant complexs were incubated in vitro for 2 days and then impanted subcu-taniously into nude mice. New bone formationre and new bone integra-tion with dental impant were evaluated by gross inspection, X - ray ex-amination , histologic observation and computerized histomorphometry 4 and 8 week after dental impantation.ResultsBy gross observation, specimen of 4 week was red and white. X -ray examination showed that there was little radiodense shadow around the dental impant . Specimen of 8week was red and had the gross ap-pearance of bone. Dental impant could be observed situating in the newly formed bone graft. X - ray examination showed that coral scaf-fold was absorbed completely. Large amount of X - ray blocking shad-ow could be observed around the dental impant Histologic examination showed that bone - like tissue formed in the pores and on the surface of CBM and in some area new bone could be observed integrating with implant in 4 week specimen. In 8 week specimen, large amount of new bone formed around the dental impant and integrated well with the im-plant. In the MCCAB, both intramembranous and cartilaginous osteo-gensis were seen with the former predominating. In the control, only slight cartilaginous osteogenesis was seen.ConclusionsTissue engineered bone graft may integrate well with titanium dental impant.
