Allogeneic BMT is an accepted treatment for many otherwise fatal of the hematopoietic system and for inherited disorders of cells of the lymphoid system and other bone marrow-derived lineages. One of the major complications of allogeneic BMT is graft-versus-host disease(GVHD), and which slower the process of allo-BMT. Immimologically, human MSCs share cell surface markers with thymic epithelium. The presence of these cell surface markers, along with the finding that MSCs are customary residents of the bone marrow microenviroment, Suggests that MSCs may play an important role in the immunoregulation provided by the bone marrow microenvironment.In this study human bone marrow mononuclear were separated by gradient centrifugation on Percoll(density 1.073g/ml).The purity of MSCs was analyzed by flow cytometry. The stromal feeder layers of irradiated MSCs were cocultureed with allogeneic T lymphocytes. The expression of the IL, IL-4, IL-10, IFN-r were evaluated after 7 days of cocultures by ELISA, and the phenotypes of the T cells were analysed by flow cytometry before and after coculture with MSCs. The purity of MSCs collected by Percoll and cultured in medium containing 10% FBS was around 95% as assessed by flow cytometry .The MSCs can sucessfully differentiate into tendonogenic cells of the and so on in vitro. CD8+ suppopulation was increased and CD25+ subset was decreased in poportion after coculture with MSCs for 1 weeks. ELISA showed that the serectionof IFN- r was upregulated., and IL-2 , IL-4 was donwregulated . The number of T lymphocytes was apparently decreased.It was concluded that the allogeneic MSCs surely affected cytokine secretion of the T lymphocytes. The secretion of the IFN- y was upregulated. It was probably associated with the change of the phenotypes of T cells. The results indicated that MSCs pretreatment might be useful in the prevention of GVHD in HLA-mismatched bone marrow transplantation and further donors for hematopietic stem cells could be selected from greater potentials.
