Thromboembolism at the vascular anastomosis site is the worst complication after transplantation of tissue and replantation of severed limb(fingers). Searching an effective method for preventing the thrombosis is still an important research topic.Ligustrazine is abstracted from Ligusticum chuanxiong Hort. Ligustrazine has extensive pharmacological function, especially for cardiovascular system and blood system. We used rabbit femoral artery-vein end-to-end anastomosis model to observe the long term patency rate and NO(nitric oxide) content and hemorhelogy were determined. ObjectiveTo study antithrombotic effects of ligustrazine in small vessels after microsurgery ,to offer experimental data for ligustrazine's clinical application into microsurgery and offer drug for improving patency rate of the site of anastomosis of microvessels. MethodRabbit femoral artery-vein end-to-end anastomosis model was set up. Both sides of femoral arteries and veins were transected respectively. Then under the microscope enlarged 10 times, the transected vessels were anastomosed with 10/0 undamage suture. After operation rabbits moved freely. 18 Japanese rabbits were divided into 3 groups randomly according to the drugs given postoperatively with 6 each. GroupA: Ligustrazine was given daily in a dose of 5mg/Kg for 15 days after operation. Group B: Heparin was given daily in a dose of 6U/Kg for 15 days after operation. Group C(control group): The similar volume of saline was given daily for 15 days after operation. On the 15th day after operation the patency of the site of the anastomosis in the vessels were measured. On the 1st 3rd 7th 15th day after operation ,the NO(nitric oxide) content and hemorheology in plasma were determined respectively. Results1. The effect, of ligustrazine on the long term patency rate.: Total 72 arteries and veins were anastomosed,which had been observed for 15days.The results showed the long term patency rates of group ligustrazine , group heparin , control group were 87.5%> 83.3%> 70.8% respectively. The long term patency rate of group ligustrazine was obviously higher than that of control group (P<0.05) .There were no difference between group ligustrazine and group heparin (P>0.05 ) .2. The effect of ligustrazine on the hemorhelogy: The hemorhelogy index of control group was obviously higher,which increased to the highest peak on the 3rd day. On the 15th day after operation the hemorhelogy indexes of three groups were no difference (P>0.05). On the 1st ,3rd ,7th day , blood viscosity, plasma viscosity and index of RBC(red blood cell) aggregation of group ligustrazine were markedly lower than those of control group (P< 0.01 ?; P<0.05 ). There were no difference between group ligustrazine and group heparin (P>0.05 ) .3. The effect of ligustrazine on NO contents: On the 1st ,3rd ,7th day ,the plasma NO content of ligustrazine were significantly higher than those of control group (P < 0.01 & P < 0.05 ) .There were no significantly difference between group ligustrazine and group heparin (P >0.05).On the 15th day, the plasma NO contents of three groups were no difference(P>0.05).ConclusionLigustrazine could reduce blood viscosity,plasma viscosity and index of RBC aggregation and increase the content of plasma NO afte the anastomosis of small vessels. Ligustrazine could improve the hypercoagulability after microsurgery. With using heparin locally during operation and using ligustrazine overall ,on the basis of successful anastomosis of the small vessels ligustrazine could prevent the vascular dangerous state and lessen the side-effect of the heparin. Ligustrazine is worthy of clinic selections of anti-thrombosis and anti-hemagglutination.
