Objective To evaluate the accuracy and clinical application of real-time fluorescence quantitative PCR (real-time FQ-PCR) in detection of Ureaplasma urealyticum(Uu) DNA in non-gonococcal urethritis and cervicitis specimens.Methods In 116 clinical specimens ,the DNA of Uu were detected by the real-time FQ-PCR, isolated by the routine culture, 20 cases of them were also identified by super-power microscope.Results In real-time FQ-PCR, the standard curve had a good agreement to serial dilutions ( 102, 103, 104,105,106 DNA copy /uL of standard template). 47 cases were positive and the positive ratio was 40.5%;in routine culture, 49 were positive and the positive ratio was 42.2%. No significant difference was found between the two methods. Compared to the culture, the sensitivity of real-time FQ-PCR was 93.9% while the specificity was 98.5% ,the agreement was 96.6%. in the 20 cases, 12 were positive by super-power microscope, 6 were positive by culture, 5 were positive by real-time FQ-PCR.Conclusion The real-time FQ- PCR is a simplejapid and accurate method for detection of Uu and it will play a crucial role in laboratory diagnosis. While the super-power microscope will give some incredible results.
