Objective: To study the effects and mechanism of NOS inhibitor on angiogenesis in the model of mice lung cancer . Methods: 14 c57 black male mice were divided into two groups (controul group and treating group),7 in each groups. Lewis lung cancer cell strain was inoculated subcutaneously to induce medel of lung cancer. The treating group mice were administrated abdomently with NOS inhibitor (5mg/kg) every day for 14 days. After 14days all mice were killed ,then took out tumor and measured the weight of tumor; couting microvessel Dencity(MVD) of tumor under light microscope. Immunohistochemistry was performed to measure vascular endothelial grouth factor(VEGF). The content of serum NO were measured by means of ELISA. Result: (1) The weight of tumor in control group and treating group were respectively 2.59±0.31 and 1.80±0.33, the weight of tumor in treating group was evidently lower than that in control group(p<0.01); (2) The MVD of tumor in control group and treating group were respectively 24.06±5.42 and 16.97±2.86, the MVD in treating group was significantly lower than that in control group(p<0.01); (3) In expression of VECF there was no difference between control group and treating group. (4) The content of NO in control group and treating group was respectively 33.26umol/L and14.56±7.13umol/L(p<0.01) ; (5) In lung cancer transfer ,under light microscope there was no difference between control group and treating group . Conclusion: (1) NOS inhibitor(L-NAME) may inhibit synthesis of inducible NO; (2) L-NAME might inhibit angiogenesis of tumor in model of mice lung cancer by means of decreasing synthesis of inducible NO; (3) L-NAME might play a role of inbibiting tumor angiogenesis by means of another route, not by inhibiting VEGF; (4)The inhibitor of angiogenesis might have a very strong potential perspective in the treating lung cancer.
