Trigeminal neuralgia (TN) is one of the common diseases of the nervous system, which can impair the life quality of sufferers. Its pathogenicity and pathologic mechanism were not decided yet, but it is widely accepted that the pathogenicity lies in peripheral nervous system and the pathologic mechanism in central nervous system, lot of factors can result in TN. Using a trigeminal neuralgia rat model, the effects of sodium valproate (VAP) or neurotropin (NTP) on the changes of asymmetric face grooming behavior were observed after formalin induced oral pain. Changes of substance P and calcitonin gene-related peptide (CGRP) also were observed in caudal spinal trigeminal nucleus (Vc) and trigeminal gangglioneure, by which the inducing pain function of SP and CGRP on TN was explored. The effects of NTP and VAP treating on TN were studied. Materials and methods1. 24 healthy Sprague-Dawley rats were randomly divided into 4 groups. Group A: infused saline plus saline; group B: formalin stimulation into the left face plus saline injection ; group C: formalin stimulation plus sodium vaproation injection; group D: formalin stimulation plus neurotropin injection. 150ul 10% formalin was injected into the left mystacial pad in groups B, C and D, at the same place 150ul saline was injected in group A, after 30 minute 10ml saline was injected in groups A and B intraperitoneally, 10ml VAP (10mg/100g) was injected in group C intraperitoneally,10ml NTP (5Nu/100g) was injected in group D intraperitoneally.2. 4 groups rats were sacrificed at 12 hour after formalin-injected. The bottom segment of brainstem and bilateral trigeminal ganglion tissue were removed and embeded in paraffin .The thick was 4um.3. Observing index: After facial injected the asymmetric face grooming behavior time was observed in 4 groups rats at 15 -25 minutes (time I), 40 -50 minutes (time II), 60 -70 minutes (timelll) by electric stop- watch.Through Nissl staining, and immunohistochemical SABC method of staining, the caudal spinal trigeminal nucleus (Vc) and trigeminal gangglioneure were observed. SP-positive and CGRP-positive neuron percentage in ganglion were detected, the percentage = (positive neuron/all neuron) * 100%. SP - positive and CGRP - positive fiber in Vc were graded: grade 1: light yellow, narrow; grade 2: yellow, wide; grade 3: deep yellow, wide.4. Analysize: the data was handled with SPSS 10.0 statistic software. The difference of groups was compared with repeated measure data analysis of variance, one-way analysis of variance and rank sum test, significant level is a =0.05.Results1. After oral injection, rats performanced uneasy, low ability of equilibrium and frequently asymmetric face grooming, after about 1 hour they behaved in low spirit, showing the tendency of sleep, which indicated the acute inflammatory neralgia rats model had been established .Rats in group A had few asymmetric facegrooming behavior, in group B, group C and D performanced typical neuralgia as above. But the symptom of rats in group C, group D relieved, at 1 hour they showed no intention of sleeping.2. The asymmetric facegrooming behavior time:Time I : group A 52.23± 13.09 seconds, group B 178.37 ± 25.14 seconds, group C 178.92 ± 34.42 seconds, group D 164.14 ±23.12 seconds; the difference between any group and group A was significant (p < 0.01). Other comparison were not significant between groups (p > 0.05) ,showed that formalin can induce obvious pain.Time II: group A 31.97 + 5.29 seconds, group B 167.77± 44.16 seconds, group C74.31± 22.70 seconds, group D 66.59± 26.79 seconds; the difference between group C or D and group B was significant (p < 0.01), suggested that both VAP and NTP can relieve the inflammary pain from 40 minutes after formalin-injection, the difference between group C and group D was not significant (p > 0.05) ,showed the effects of two medicine had no obviously difference.Timelll: group A 11.06± 6.12 seconds, group B 78.65± 13.48 seconds, group C 33.77± 24.96 seconds, group D 22.93± 18.5...
