Oxytropis Kansuensis Bunge is one of toxic whin category plants, which distributes abroad, has high nutrient value and toxicity, and seriously threatens the local stockbreading and causes massive economic losing. Now, the research of whin category plants in prevention or killing off and utilization has made good progress, but its officinal research is not in-depth and not systemic. The experiment aims at selecting the anti-tumor component or sect from Oxytropis kansuensis and lays a foundation for exploiting Oxytropis Kansuensis to a new and effective anti-tumor herbal medicine.The experiment generally includes the following three contents.I.We applied traditional phytochemistry separate method to get four different polarity parts, namely petroleum ether part, chloroform part, acetic ester part and water part, we also separated alkaloid part and non-alkaloid part from Oxytropis Kansuensis, based on research of alkaloid from sibling plants of Oxytropis Kansuensis. All these separated parts will be done selecting experiment on anti-tumor.2.After chemistry composition and mice toxic previous experiment, we picked up acetic ester part, water part, alkaloid and non-alkaloid part to carry out the anti-tumor experiment on mice bearing sarcomaisoand H22. According to result, alkaloid part can be considered as the effective anti-tumor sect of Oxytropis Kansuensis.3. In order to study the prevention role and the possible mechanism of alkaloid from Oxytropis Kansuensis in mice bearing sarcoma^o, the experiment observed changes of malignant phenotype of mice sarcomaiso cell by light microscope and studied the expression of proliferating cell nuclear antigen (PCNA) and mutant p53 protein with immunohistochemistry.The experiment has achieved the following results.l.The research on histology of Siso cell treated with alkaloid indicates that alkaloid of Oxytropis Kansuensis has direct effect on S180 cell. The changes of malignant phenotype of mice S180 cell were observed by light microscope. The result is that alkaloid from Oxytropis Kansuensis can inhibit the malignant phenotype of mice sarcoma, cell such as heteromorphism, heteropyknosis and increase of nucleus/plasma ratio. The alkaloid can also cause tumor cell putrescence such as nucleus atrophy nucleus disintegrating and nucleus dissolving. Remaining tumor cell is regular circle or ellipse.2. The expression of proliferating cell nuclear antigen (PCNA) and mutant p53 protein were studied with immunohistochemistry. The result is that the expression of PCNA and mutant p53 protein compared with negative control group, two groups both are reduced ( P<0. 05 or P<0. 01). It suggests that alkaloid from Oxytropis Kansuensis has a good role on inhibition sarcoma, of mice, this role is associated with the reduction of PCNA and p53 protein.3. The comparing results of rate refraining from tumor growing in groups: all of the rate refraining from tumor growing in non-alkaloid part groups are less than 30%. According to the evaluating criterion of selecting anti- tumor medicine in vivo, it is no effective. To some extent, acetic ester part and water part both have effect on mice bearing S180 and H22, but the effects of them are both inferior to that of alkaloid part.4. The effect of every treat on immune organs and weight of mice bearing S180 and H22: the weight analysis to thymus and spleen indicates that every treat has no obvious effect on immune organs. Compared with control, the rate of gaining body-weight has no difference (P>Q. 05).The conclusion of the test:1. For the first time, the experiment carried out the systematic selection of anti-tumor composition from Oxytropis kansuensis according to the methods of the herbal medicine development.2. The experiment confirmed that the alkaloid part was effective anti-tumor part of Oxytropis Kansuensis. Owing to a certain amount alkaloid distributing in acetic ester part and water part, they present inferior effect on anti-tumor.
