When interferon was discovered in 1957, it was hailed as a significant antiviral agent. In the late 1970s, interferon made a big splash as a symbol of recombinant gene technology and the medical breakthroughs it would bring. The natural antiviral and antitumor activity of interferon alpha and beta promoted investigation for therapeutic use. Interferon therapy has been successfully implemented for skin malignancies, viral infections, kaposi sarcoma, multiple sclerosis, leukima, et al.Human interferon alpha-2b(huIFN- a 2b) gene was inserted into bombyx mori baculovirus transfer vector pBacPAK.8 and cotransfected with lineared DNA of BacPAK6 virus into BmN cells. The homologus recombination occurred inside the cells, and then the recombination virus Bac-IFN a 2b, identified by southern, was expressed. The BmN cells the fifth instars were infected by the recombination virus Bac-IFN a 2b; And the expression product of protein was run in the SDS-PAGE, and its immunoreactivity was determined by using ELIS A and Western blotting. The bio-activity of protein product was determited by using the method of cytopathic effect inhibition assay (CPE). The expression activity achieved the highest level at 72nd hour in BmN cells (4.1 × 106IU) and at 144nd hour in larvae (4.0× 106IU). And the expression product can alleviate the symptoms of liver trauma of ICR mouse.In conclusion, we could obtain lots of active alpha-2b interferon peptide by BEVS and lay a foundation for clinical application research of interferon.
