The silkworm was recognized as curiosity to treat illness and nourishment for a long time. There was recordation that the silkworm could treat diabetes. To analyze the factors of the treatment of silkworm to diabetes, there were two major aspects, one was that there were a lot of flavonoid contents in the silkworm, which could boost the insulin B cell excrete insulin, reduced the blood sugar and serum cholesterin, improved the endurtion of sugar, contradicted the improving sugar function of adrenalin .So it could bring the function of reducing the blood sugar. In this experiment, we measured the flavonoid contents with spectrophotometry and the DNJ content with reversed-phase high-performance liquid chromatography. This paper analyzed the factors to affect the content of flavonoid and DNJ in the silkworm body. Based on this, we could provide theoretical basis to develop medicine to cure diabetes.After qualitative analysis with the methods of Polyacrylamide gel film chromatographic analysis and muriatic acid-zinc, we discovered that the silkworm body fluid turned into red after the reaction of muriatic acid-zinc. It could produce kelly fluorescence under the ultraviolet lamp after it was suffocated with ammonia steam and treated with 5% three-chlorination aluminium. The result indicated that there was flavonoid in the body of silkworm. The flavonoid content could be exactly and stably mensurated with ultraviolet spectrophotometer after it was treated with the process: it was sifted with 80 item and mixed with grain alcohol at the proportion of 1:30.then with the circumfluence for 4 hours at the temperature 60癈 for three times.There was rich flavonoid in the silkworm larval body, and the content of flavonoid obvious different with variety, gender and the phrase of growth. In the 25 kindes of laval bodies, the flavonoid contents was 11.107~34.740 mg/g, and the content of flavonoid in the female silkworm is higher thanthat in male body. For the same kind and different phrases, the content of flavonoid was highest in the third day of fifth instar.This experiment was based on FMOC-C1 and DNJ compound, DNJ-FMOC-C1 had fluorescence, so it could be measured by fluorescence detector, so we could measured the content of DNJ with reversed-phase high-performance liquid chromatography. This method was very precise, stable and effective.There was rich DNJ in the silkworm larval body and the content of it was 0.448%, which was higher than mulberry leaves and the content of DNJ was obvious different with variety, gender and the phrase of growth. In the 25 kinds of silkworm, the content of DNJ was 0.26-0.53%, and the content of DNJ in the male silkworm was higher than that in female body. In the ages of 1 to 5, the content of DNJ was highest in the flourishing age of third instar and lowest in the flourishing age of the fifth instar. In the different phrases of the fifth instar, the content of DNJ in the third day was highest, and the DNJ was not found in the silkworm chrysalis, ant silkworm larva and silkworm pupae.
