The Dynamical Detections And Their Significances Of Interleukin-18(IL-18) And NO(Nitric Oxide) During Experimental Mice Acute Hepatic Injury Induced With Concanavalin-A And The Exploration Of Mechanism Of PHGF's Protection Liver

Objective To investigate the relations between interleukin-18 (IL-18) ,nitric oxide(NO) and acute hepatic injury and the relations between IL-18 and NO by dynamical checking and measuring the levels of IL-18 and NO during acute hepatic injury at different time,and to observe the effects of PHGF(promoting hepatocyte growth factor) on production of serum IL-18 and NO and investigate the mechanism of PHGF which protect liver tissures during mice acute hepatic injury. Methods mice were randomized and divided by threeg groups, control group and test group and protection group.and the blood samples of three groups mice which were handled with reagents or medicine were collected at 2h , 4h, 8h, 24h, hepatic part tissures of 10 mice in every group were resected to be made homogenate at every different time point,then we measured the level IL-18 and NO in both serum and liver homogenate and alanine aminotransferase(ALT) levelonly in serum. At last, hepatic pathological changes were checked and hepatic injury degrees were ranked by optical microscope. Results IL-18 level both in serum and in liver tissure at different time points were signiticantly higher and ALT level in serum significantly higher in test group than in control group. Serum NO level in test group significantly lower than in control group at 2h time point, but significantly higher at 4h and 8h time point,NO has a both significantly negative(2h) and positive correlation(4h and 8h) with acute hepatic injury. IL18' s and NO' s values were Maximal in test group both in serum and in liver tissure at 4h,and both of them have a significant correlation with acute hepatic injury.IL-18 level in liver homogenate is significantly higher but NO significantly lower than in serum,IL-18 has a positive correlation with NO in test group at 4h.IL-18 level in protection group is no significantly difference but NO significantly lower than in test group,at the same time, ALT level significantly lower in protection group,too. Conclusion IL-18 may play a key role during acute hepatic injury maybe it is because that IL-18 can make hepatic cell product NO and all kinds of cell factors which can make liver damaged. We have found out for the first time NO makes the dual effects during acute hepatic injury. A significant correlation relation exists between IL-18 and NO,and both of them can cause hepatic cell injured synergetically. When diagnosing the serious degree of liver damage We think determining IL-18 in liver tissure homogenate is more sensitive than in serum,on the contrary, determining NO in serum is moresensitive.We have testified PHGF made no effect on IL-18 for the first time,but it can decrease NO significantly,which suggest that PHGF protect liver from damage by reducing NO.our study results not only provide experiment basis for us to research and develop new medicines which can protect liver from damage,but also does it establish theory foundation for us to prevent and treat all types of hepatitis.
