Direct Gene Transfer Of Naked DNA Encoding Vascular Endothelial Growth Factor Into Hypoxia Ischemia Damage Brain Of Newborn Rat

Hypoxis ischemic encephalopathy(HIE) is a kind of newborn disease,which can lead to nervous system dysfunction.Hypoxic and ischemic is the most important reason.lt is the key piont for the prognosis to improve Anoxia of brain and reduce brain damage .Vascular endothelial growth factor (VEGF),also known as vascular permeability factor (VPF),can works on endothelial cells,peticuliarly to induce endothelial cells mitosis,proliferation and migration. VEGF also is a direct neuroprofective factor for neurocyte and endothelial cells.To explore the function exogenesis VEGFgene in the HIBD of newborn rats ,VEGF eukaryotic expression plasmid (pCDNA3.1/rVEGF120)was constructed. pCDNA3.1/rVEGF120 was injected into hypoxia-ischemic nexbom rat brain following the HIBD.Methods:VEGF eukaryotic expression plasmid (pCDNA3.1l/rVEGF120) was constructed by cloning rat VEGF 120 cDNA into eukaryotic expression vector pCDNA3.1.The HIBD model was established with seven days old SD rats,and all rats were diveded into twogroups at random:central group 18 rats(every rat model was injected pCDNA3.1100ug immediately after hypoxia-ischemic,then raised seven days) and therpy group 18rats (every rat model was injected pCDNA3.1/rVEGF120 100ug immediately after hypoxia-ischemic.then raised seven days).they were killed,and brain were removed at once .immunohistochemistry.dot blotting and flow cytometer(FCM) were used to investigate VEGF gene expression and biological effect.Result:High VEGF protein was detect in neurons ,astrocytes and vescular endothelial cells in immunohistochemistry . Dot blotting detected high VEGF protein , too. There was a significant decreas in the ratio of apoptosis cell.Conclusion:1.the topical VEGF gene application can transfect the ischemic brain tissue and express VEGF protein .the VEGF gene product can reduce apoptosis .
