Influence On Apoptosis Of Transferring Vascular Endothelial Growth Factor Gene Into Hypoxia Ischemia Reperfusion Damage Brain Of Newborn Rat

Objective:Vascular endothelial growth factor(VEGF)is idiosyncratic mitosis ortho for the endothelium cell of blood vessel, it can stimulate angiogenesis of some tissue such as skeletal muscles,myocardium and brain and decrease the apoptosis of neurocyte. To observe the influence on genes correlated with apoptosis after transferring VEGF gene into hypoxia ischemia--reperfusion damage brain of newborn rat by eukaryotic expressing vector—pcDNA3.1/rVEGF120 in order to investigate possible mechanism and feasibility of VEGF gene therapy of newborn rat for hypoxia ischemia--reperfusion brain damage.Method:To transfer VEGF gene into brains of seven-day-old SD rats simulated hypoxia ischemia-reperfusion damage by eukaryotic expressing vector-pcDNA3.1/rVEGF120 , expression phases of apoptosis correlation genes (bcl-2,bax and fas) were detected by immunohistochemistry (IMH) and Hoechst dye.Result:Expression of VEGF gene starts up the 3rd days after model of newborn rats hypoxia ischemia-reperfusion brain damage has been made , protein of VEGF gene can be observed and increased gradually from the 3rd day to the 14th day by IMH dye in therapy group. The number of apoptosis neurocytes have been decreased in therapy group. Protein of anti-apoptosis gene, bcl-2 ,is observed from the first day to the third day by IMH dye in therapy group and is up regulated .And that of the promote apoptosis gene ,bax and fas, are down regulated compared with control grounp. Conclusion:VEGF gene injected directly into rat brain express its protein efficiently and continuously at least for tow weeks. And it can decrease neurocytes apoptosis perhaps by regulating the expression of genes correlated with apoptosis.
