Objective:To investigate mechanism and curative effect of Electroacupuncture in Dazhui and Baihui of vascular dementia(VD).Methord:Wistar rats were randomly divided into control group, model group, electroacupuncture(EA) group and hydergine group. Rat model of VD was established by a modified Pulsinelli's 4-vessel occlusion(4-VO) method. Y-maze was used to observe, the changes of capability of learning and memory. After the treament, Wistar rats were killed and the content of somatostation(SS) and the activity of acetylcholinesterase (ACHE) in rats' hippocampus were measured with radioimmunity method and colorimetric method . Ultrastructural changes of neurons in hippocampus were observed with transmission eletron microscope.Results: The model rats of VD needed more Y-maze test times than the control rats(p<0. 01), showing severe impairment of memory. The SS and ACHE contents of hippocampus in model rats were decreased significantly in comprison with the control group(p<0.01).The ultrastructure of neurons in hippocampus were altered obviously and damaged severely; In the EA group and hydergine group , the Y-maze tests times reduced significantly in comprison with the model group (p<0. 01), the times were not different from the control group rats , showing the improvement of learning and memory ability. Compared with the model group , the content of SS and ACHE in rat hippocampus were raised significantly in EA and hyderine group(p<0. 01, p<0.05). There is no difference between the EA and hyderine group on the content of SS and the activity of ACHE in rat hippocampus . The damages of the neurons of hippocampus in the EA and hyderine group were obviously slight in comprison,with the model group .conclusion:EA could reduce the damage of neurons in hippocampus , and then raised activity of AchE and content of SS, these are conducive to improve learning and memory ability of VD model rats. The mechanism of treating VD rat by EA could have relations with regulation of cholinergic neurotransmitters and neural peptid, these offered the experimental evidence of EA treatment on VD.
