Vascular dementia caused by cerebrovascular disease(including ischemic encephalopathy, hemorrhagic encephalopathy and chronic hypoxic-ischemic encephalopathy) is a condition of impaired intellingence and cognition.With the aging tide coming,vascular dementia becomes a predominant disease that harm old people'health.So how to prevent and treat VD is a social problem and a task of medical worker.Purpose:Determine the ability of learning and memory of rats by behavior experiment and the expression of Super oxide dismutase(SOD),Malondialdehyde and acetylcholines-terase (AchE) in hippocampus tissue of rats and detect the apoptosis of neurons in hippocampus tissue. Explore the mechanism of action that Rhodiola improved the intelligence of vascular dementia rats.Provide the experimental basis for Rhodiola clinically.Methods:Occlusion of bilateral common carotid arteries was applied on health male Wister rats to make chronic hypoperfused brain,which could establish a model of vascular dementia. Every group is eight.After operation,Rhodiola group and Nimodipine group were intragastrically given to rats by a dose of 5g/kg/d and 20mg/kg/d respectively.While the rats in sham operation control group and model group were intragastically given the same volume distilled water.We evaluate the effect of action that Rhodiola improved the intelligence of vascular dementia rats by detecting the SOD activity,MDA level,AchE activity and neuron apoptosis in hippocampus tissue.Results:1 Rhodiola can improve learning and memory abilities of VD rats in the Morris water maze.There are significant differences between Rhodiola group and model group(P<0.05). 2 Rhodiola can increase the activity of SOD and reduce the content of MDA and activity of AchE in the hippocampus of VD rats.There are significant differences between the Rhodiola group and the model group(P<0.05).3 Rhodiola inhibits neuron apoptosis in hippocampus tissue of rats,there are significant differences between the Rhodiola group and the model group (P<0.05).Conclusions1.Rhodiola can significantly improve spatial learning and memory impairment in VD rats, which suggest that Rhodiola can prevent and treat VD. 2.Rhodiola can improve SOD activity and reduce the content of MDA in the hippocampus of VD rats,suggesting that anti-lipid peroxidation is one of the mechanisms that Rhodiola improves spatial learning and memory impairment in VD rats.3.Rhodiola can inhibit AchE activity in the hippocampus of VD rats,suggesting that anti-cholinergic neuron injury is one of the mechanisms that Rhodiola improves spatial learning and memory impairment in VD rats.4.Rhodiola can significantly reduce neuron apoptosis in hippocampus tissue of VD rats, suggesting that Rhodiola can protect neuronal cells and inhibit apoptosis.There are relationships between the above effect and improvement of spatial learning and memory impairment in VD rats.
