Objective: To e×plore the effect of c-fos and c-jun antisense gene recombinants transfection on protection of cardiomyocytes following hypoxia and burn sera treatment.Methods: Burn sera was collected from wistar rat with 30%Ⅲ°TBSA, cardiomyocytes cultured in the mixture gas containning 1% O2 was used as hypoxia model. The c-fos and c-jun antisense gene recombinants were constructed. Neonatal Wistar rat cardiomyocytes were cultured in vitro with hypoxia and burn sera treatment. c-fos and c-jun antisense gene recombinants were transfected into the cultured cardiomyocytes. Expressions of c-fos and c-jun mRNA were determined with RT-PCR. Morphological changes of cardiomyocytes were observed under light microscope and electronic- microscope. Expressions of c-fos and c-jun protein, troponin, β-Tubulin in cardiomyocytes were assayed with Western blot in the transfected and non-transfected groups.Result: Under light microscope, the morphology of cardiomyocytes in the group only adding the burn sera and hypoxia (the non-transfected group) changed explicitly. Disarrangement of myocardial cytoskeleton net, fragmentation of cytoskeletons and formation of granules distributing around nucleus were observed. The filaments in cytoplasma connected into nets, distributing radially from nucleus to outsides. Dissolvation of the strucrure of microtubulin and frangmentation in the non-transfected group were observed under electronic microscope. However, these changes were not so obvious in the transfected cardiomyocytes, the structure of microtubulin almost remained normal. Results of RT-PCR showed that the expressions of c-fos and c-jun mRNA increased significantly in the non-transfected group. But after transfecting c-fos and c-jun antisense gene recombinants, the expressions of c-fos and c-jun mRNA decreased significantly compared with non-transfected cardiomyocytes. Results of Western blot show that the expressions of c-fos and c-jun protein in the transfected group decreased apparently compared with the non-transfected group, but the expressions of β- Tubulin and troponin increased significantly in the transfected group. Conclusion: The burn sera and hypoxia can cause the injury of cardiomyocytes,transfection of the c-fos, c-jun antisense gene recombinants may reduce the injury of cardiomyocytes. The results suggest that the c-fos and c-jun antisense gene recombinants transfection have the protective effect on cardiomyocytes exposed under burn sera and hypoxia.
