| Objective To observe the effect of atorvastation preconditioning on the expressions of matrix metalloproteinases-2(MMP-2) and cardiomyocyte apoptosis after hypoxia/rexygen injury on rat cardiomyocyte.Methods To culture cardiomyocyte cells and underwent randomly into1) received no intervention (contral group,n=6),2)exposed to hypoxia and reoxygen(H/R group,n=6), put cardiomyocyte cells into obdurate container,then pump the air from it and input air (95%N2+5%CO2)for20seconds, then get them out of the container and keep for6hours (37℃,95%air,5%CO2);and after that put them into the cell container for2hours.3)received0.1μmol/L atorvas-tation (low dose) before hypoxia and reoxygen (H/R+L group,n=6),4) received1μmol/L atorvastation (high dose) before hypoxia and reoxygen (H/R+H group,n=6). atorvas-tation was matched different does in DMEM.MMP-2level was test with enzyme linked immunosorbent assay (ELISA) and cardiomyocyte apoptosis was detected with flow cytometry.Results The level of MMP-2and cardiomyocyte apoptosis of HR group were significantly higher than control group (20.324±10.152VS128.365±51.61;(4.6±7.24)%VS (46±6.557)%P<0.05);The level of MMP-2and cardiomyocyte apoptosis of HR group and H/R+L group were no significant differences;(20.324±10.152Vs126.263±35.225;(46±6.557)%Vs (22±5.568)%P>0.05); H/R+H group were lower than H/R+L group (71.137±11.157Vs126.263±35.225,((20±4.02)%Vs (22±5.568)%p<0.05)Conclusion High dose atorvastation can protect rat cardiomyocyte from hypoxia/reoxygen induced high MMP-2level and apoptosis. |
