Objective To study the effects and mechanism of Naotaifang extract preconditioning on the cerebral ischemia-reperfusion injury in gerbil.Methods: Cerebral ischemia-reperfusion models were induced by bilateral carotid occlution in gerbils. 130 gerbils were randomly divided into the sham operation group, the cerebral ischemia-reperfusion injury group(IR group), the cerebral ischemia-preconditioning group (IPC group), and Naotaifang extract preconditioning group (NPC group). According to the time of the ischemia-reperfusion (24,48,72h and 7d), IR group, IPC group, and NPC group were divided into 4 subgroups. Adenosine(ADO) contents and nitric oxide(NO) in the serum, the water contents and Evan's blue(EB) in the brain tissues were measured. The number of survival neuron in the gerbil hippocampal CA1 region and the ultrastructure changes of the hippocampal CA1 region were observed.Results: 1. Compared with IR group at the same time-shape of reperfusion, The water contents and Evan's blue of brain tissue in IPC group and NPC group were reduced significantly (P<0.05 or 0.01),the number of survival neuron in the gerbil hippocampal CA1 region IPC group and NPC group was increased significantly(P<0.05); and the ADO contents and NO in the serum of IPC group at the 48th , 72th h and NPC group were enhanced significantly (P<0.05). 2. After treatment. The water contents and EB in thebrain tissues arrived the peak at the 72th h of reperfusion in each group.The ADO contents in the serum arrived the peak at the 7 day of reperfusion in each group. The number of survival neuron in the gerbil hippocampal CA1 region and the NO contents in the serum arrived the lowest at the 7th day in the each group. 3. The number of survival neuron in the gerbil hippocampal CA1 region and the water contents had no difference between NPC group and IPC group (P>0.05). There were significant difference between NPC and IPC in the levels of NO and ADO in the serum and the contents of Evan's blue in the brain tissues(P<0.05).Conclusion: Naotaifang extract preconditioning inhibits the cerebral ischemia-reperfusions injury in gerbil.
