Objective : The research of glycine on chronic hepatic diseases and it'smechanism.Methods : Part I : 48 patients with chronic hepatic disease were equally divided into 2 groups. Among them ,24 patients were given basic treatment as control group. 24 patients were treated to basic medicine and glycine as therapy group. Clinical study has been analyzed. The plasma has been collected for measure of ALT, AST, TBiL, PA, ET ,TNF- α and IL-6 levels, etc. Part II: The chronic liver disease's model of wistar rats was established by complex pathogens which have been fed with control diet and treatment group which have been fed with control diet adding 5% glycine at the same time. The samples has been taken on 4 and 8 weeks end. Hepatic tissue has been collected for measure the expression of CD14mRNA by reverse transcriptase-Polymerase chain reaction (RT-PCR). Results : Part I : (1) All data showed that the therapy group getsignificantly improved after treatment(P<0.05). However the control group only ALT and PA show significantly change(P<0.05). (2) AST, PA, ET, TNF-α and IL-6 show significantly difference in the therapy group compared with control group(P<0.05). ALT and TBiL were lower in therapy group than both in control group. But statistics has no significantly difference. (3) The clinical effects of therapy group become better than control group. (4) Close relationships has been found between ET, ALT, AST, TBiL, PA, TNF- α and IL-6. Part II :The CD14mRNA expression of hepatic tissue from chronic liver disease's rats which are fed with containing glycine diet was weaker than the control group, and the CD14mRNA expression on 8 weeks end chronic liver disease's rats feeding with containing glycine diet was the weakest.Conclusion : 1. Glycine can resist liver injury , improve liver function , elevate ET , TNF- a , IL-6 level and remarkably decrease IETM effect. 2. Glycine can remarkably block the CD14mRNA expression of hepatic tissue from chronic liver disease's rats.
