Part OneThe Effects of ZhiHuang Decoction on levels of TNF-αand endotoxin in serum in the rats with acute alcoholic hepatic damageObjectTo study the effects of Zhihuang Decoction (ZHF) on acute alcoholic hepatic damage and its mechanism,we investigated the morphological changes of hepatic tissue and the levels of TNF-αand endotoxin in serum of the acute alcoholic hepatic damaged rats treated with ZHF.MethodsThirty wistar rats were divided into treatment group, model group and normal group, Each group had 10 rats. After 10 days of making the model, the changes of liver morphology were observed with HE staining, the expression of TNF-αprotein in liver was tested by immunohistochemistry, and the endotoxin protein and ALT in plasma was assayed by biochemistry. Result We observed the histological and morphologic changes in hepatic tissue. In the normal group, hepatocyte was arranged in order, the cellular plasm was symmetrical, the cellular contour was clearly, and the structure of hepatic cord's reticular fibre was integrated; In the model group, hepatocyte arranged in disorder, the cellular plasm was like balloon degeneration, the hepatocyte was fatty degeneration; Compared with the model group, hepatocyte arrange was better, and hepatocyte contour was clearer in the treated group. The level of endotoxin in the treated group was higher than that in the normal group (P<0.05), and it was significantly higher than that in the model group (p<0.01). Compared with the normal group, the content of TNF-αand ALT also were higher in the treated group (p<0.01), and they were significantly less than those in the model group (p<0.01). The endotoxin of plasma was positively correlated to the content of TNF-α(r=0.993, p<0.01).ConclusionZHF can significantly decrease the TNF-αand endotoxin in serum of the rats with acute alcoholic hepatic damage. Each group had 10 rats. We made the model of ADH by perfusing with Liber-Decarli liquid food added with Ferrous Sulfate. The changes of hepatic morphology was observed with HE staining, the expression of CD14 and IL-1 in liver was tested by immunohistochemistry. The TG and TC in serum were assayed by biochemistry.ResultWe observed the histological and morphologic changes in hepatic tissue. In the normal group, hepatocyte arranged in order, the cellular plasm was symmetrical, the cellular contour was clear, the structure of hepatic cord's reticular fibre was complete. In the model group, hepatocyte arranged in disorder, the cellular plasm was like balloon degeneration, hepatocyte had fatty degeneration and acidophilic necrosis. Compared with the model group, contour of hepatocyte was clearer, and hepatocyte arranged was better in the treated group. Compared with the treated group, contour of hepatocyte was more vague, and histology of hepatocyte had changed worse in the group with abstinence from alcoholic. Compared with the model group, the TG and TC in serum declined more obviously in the treated group (P<0.01, P<0.05). Compared with the group with abstinence from alcoholic, the TC also declined more in the treated group (P<0.05). The CD14 and IL-8 of the treated group were higher than that of the normal group (P<0.05, p<0.01), and they were less than that of the model group and the group with abstinence from alcoholic.ConclusionZHF can inhibit significantly the increase of CD14 and IL-8 in hepatic tissue, and it can improve the hepatic morphologic pathological changes in rats with ALD.
