| objective To evaluate the role of NF-кB and ICAM-1 in course of airway inflammation of smoking rat by observing bronchial mucosa inflammation and corresponding expression of NF-кB and ICAM-1 in airway epithelium cell. Method Rat smoking device was performed byself. 39 rats were divided into 3 groups :control group, smoking group of month 1, smoking group of month 3, 13 rats in each group. To compare airway mucosa pathologic change of 3 groups and expression of NF-кB(p65) and ICAM-1 protein were measured by immuno histochemistry stain respectly. In addition, Expression of ICAM-1 mRNA was investigated by in situ hybridization.Result (1)To compare with control group ,there was obvious inflammation cell infiltration in airway mucosa of smoking group rats of month 1, and its mucosal membrane hadn' t kept integrated and necrosis could be found . But significant hyperplasia of gland and smooth musle layerwere not seen. There was typical pathologic change of chronic bronchitis and COPD in smoking group of month 3. (2) The levels of NF-кB(p65) in nucleus of bronchiole epithelium cell in smoking group of month 1 and month 3 were both higher than those in control group , p<0. 01. (3) Expression of ICAM-1 mRNA and ICAM-1 protein in bronchiole and bronchi epithelium cell in smoking group rats of month 1 and month 3 were increased significantly comparing with those in control group, p<0.01. Besides, those in smoking group of month 3 were higher than those in smoking group of month 1, p<0. 05 (4) The levels of ICAM-1 mRNA and ICAM-1 protein in bronchiole epithelium were signi- ficantly higher than those in bronchi epithelium, p<0.01. (5)The percentage of NF-кB nuclear staining positive cells correlated with the level of ICAM-1 protein in smoking group of month 1,but there weren' t correlation between them in smoking group of month 3.Conclusion Smoking can induce airway inflammation process while injuring lung tissues directly .
