Objective: To observe the reversion effect of Tetrandrine tested rat Pulmonary fibrosis organization pathology, testing calcium contest and CaM activity in the fibrosis lung organization, analysing the calcium content and the intrinsic relationship between the CaM activity and the forming of fibrosis lung, inquiring into the mechanism Tetrandrine anti- rat lung fibrosis.Methods: Establishing the Pulmonary fibrosis model of rat in accordance with method of Snidert to perfus stomach with tetrandrine(30mg.kg-1 and 60mg. kg-1),they will be killed separately after the model has been made for 7, 14 and 28 days, each time kill 8 rats of each group, the normal group should all be killed after the model has been made for 28 days. After turning on the chest, fetch the left lung organization, using the atomic absorption spectrophotometer with 9003 form and cyclic nucleotide phosphodiesterase to measure calcium content and Cam activity;fetch the right lobe of the lung, judge the degree of Airsacculitis and lung fibrosis by the Szapiel method below the optics microscope; ratio analysis; Using the computer pictures to analyse and compare the average surface area of each pulmonary alveolus, Airsacculitis and lung fibrosis. Deal with and analyse the data by statistical method.Results: Pathology histology is watched: lung organization of control group is normal , the other groups all express different degree Airsacculitis, and till 28 days, pulmonary interstitial fibrosis among the lung all had happened. The degree of airsacculitis and pulmonary interstitial fibrosis with Tet's two dose groups all obviously relieve as compared with the control group, the lung organization morphologyquantitative analysis; The different surface area of different period dyes district in pattern group all increase more than the normal group. Tet's two dose groups surface area of no dying district obviously increases. The tint area and fuscous area of different period obviouly decrease (p<0.05), that illustrate the regular pulmonary alveolus cavity area adds, the pulmonary interstitial fibrosis degree eases. Different sereies of fibrosis lung organition calcium content of model groups rat obviously increase compared to the control groups. Tet two does groups of each stage lung organisation calcium content all obviously decrease compared to model groups, and the difference possesses significance(p<0. 01).Conclusion: Tetrandrine can decrease the inflammation of pulmonary alveolus which induced by bleomycin A5, antagonize the shaping of lung fibrosis and decrease the calcium content and activity of CaM. The mechanisms with protecting the Pulmonary fibrosis may be concerned with the bloking function of the calcium passage and CaM.
