| Objective: To study the injury of injecting morphine on the function of hypothalamic-pituitary-ovarian of female rat. Methods: The model rat was made by injecting morphine. The serum concentrations of FSH, LH, E2 and P were measured by radio- immunoassay after 10 weeks .The change of ER and β-EP on hypothalamic-pituitary-ovarian were observed by using immunohistochemistry technique and image analysis system. And using in situ hybridization express μ- opoide receptor mRNA. Results: (1) It was showed that the estrous cycle of 60% rat was disturbanced when injecting morpine for 7 weeks,and that of 16.1% rat was depressed after 10 weeks. (2) The serum concentrations of FSH,LH, E2 and P have significantly decreased (P<0.05, P<0.05, P<0.01, P<0.05) when the function was disordered by injecting morpine. (3)The immunoe-activity of ER in hypothalamic, pituitary ovarian.is lower than control.Conclusion: Injecting morphine long term will injury the function of hypothalamic-pituitary-ovarian axis. (4) It was showed that injecting morphine long time can reduce the positive substance of β-EP on hypothalamic pituitary and ovariana. But the express of μ-opoide receptor mRNA was increased significanterly than control.Conclusions: Injecting morphine long term will injury the function of hypothalamic-pituitary-ovarian axis. It suggests that morphine can injury on hypothalamic pituitary and ovariana and reduce the secretion of β-EP by combining with μ - opoide receptor. |
