Objective:To observe the effect of Yangjing Zhongyu decoction on AKT/m TOR signaling pathway in cisplatin-induced premature ovarian failure(POF)rats,and to explore its mechanism of improving ovarian function.Methods:42 rats were random Ly divided into blank group(n=7)and model group(n=35).The blank group was not treated,and the model group was intraperitoneally injected with cisplatin solution 4.0mg/kg,once a week,twice in all.After modeling,the model group was random Ly divided into model group,positive control group and low,medium and high dose groups of traditional Chinese medicine,with 7 rats in each group.Each administration group was given corresponding drugs for 21 days.The contents of E2,FSH,AMH and Gn RH in serum were detected by ELISA.The pathological morphology of rat ovarian tissues was observed by HE staining.The protein expressions of AKT,m TOR and Gn RHR in rat ovarian tissues were detected by IMH.The m RNA relative expression levels of AKT,m TOR and Gn RHR in ovarian tissues of rats were detected by real-time fluorescence quantification.Results:Compared with the blank group,the serum E2 and AMH levels of the model group were decreased,the FSH and Gn RH levels were increased(P<0.01),and the relative protein and m RNA expressions of AKT,m TOR and Gn RHR in ovarian tissues were decreased(P<0.01).Compared with the model group,the number of mature follicles in the ovary of rats in each administration group was increased,the number of atresia follicles was decreased,and the ovarian index was increased(P<0.05,P<0.01).The serum E2 and AMH levels of rats in the Yangjing Zhongyu Decoction high-dose group were increased,and the levels of FSH and Gn RH were decreased(P<0.01).The relative protein and m RNA expressions of AKT,m TOR and Gn RHR in ovarian tissues increased(P<0.05,P<0.01).Conclusion:Yangjing Zhongyu Decoction can improve the pathological state and ovarian reserve function of POF rats,and its mechanism may be related to regulating the function state of hypothalamic-pituitary-ovarian axis,thereby regulating the hormone level of rats,improving endocrine disorder,reducing granulosa cell apoptosis,and promoting follicle development.
