Objective: To investigate the effect of Genistein on the proliferation of hepatocellular carcinoma cell SMMC-7721 and its possible mechanism involved.Method:The effect of Genistein on human hepatocellular carcinoma cell SMMC-7721 was studied by using MTT. Flow cytometry(FCM) analysis was carried out to quantify the expression of tumor-related gene P53 , c-myc, CyclinD1.Results:(1)Genistein(2 g/ml-32 g/ml) had a dose-dependent antiprolifertion effect on SMMC-7721 cells,compared with control group. After treatment with 2 g/ml Genistein for 48 , 72 hours, the inhibiting rate reached 20.04%, 29.59% ;32ug/ml Genistein's inhibiting rate reached 98.43%, 97.59% respectively. (2)Genisteinalso had a time-dependent antiprolifertion effect on SMMC-7721 cells. After treatment with 8g/ml Genistein for 24, 48 , 72 hours, the inhibiting rate reached 14.22% , 43.21%, 47.91%;for the same time the inhibiting rate that group was treated with 16 g/ml Genistein reached 21.08% , 64.58% , 94.50% respectively. Genistein can inhibit the proliferation of the SMMC-7721 cell line in dose-dependent and time-dependent manner.(3)FCM revealed that P53 , c-myc and CyclinD1 were all down-regulated in dose-dependent manner.Conclusion: Experiment in vitro showed that Genistein could inhibit the proliferation of the SMMC-7721 cell line , the antitumor mechanism may be related to the decreased protein-expression of P53, c-myc and CyclinDl. Genistein may have a potential value in clinically treating hepatocellular carcinoma.
