Objective:TO evaluate the effect of blood vessel activity materials,cytokine,lung tissue pathological change in rabbits pulmonary embolism ischemical reperfusion injury and the application value of XingDing preventionMethods:Twenty-seven New Zealand big ear rabbits were obstructed the left lung artery by Berman sacculus catheter then it was divided into two groups randomly at 2h after pulmonary embolism operation: ⑴,IR-treatment group(n=13);⑵,IR- control group(n=14).IR-treatment group received 12ml/kg XingDing (Egb761 4.1mg in XingDing per milliliter) which was pumped into body from left-sidedness femoral vein in one hour,immediately the sacculus was put out the gas to result in reperfusion. We replaced Egb761 injection with saline in IR control group.another 11 rabbits were as sham operation group. We detected TNF and ET by radio-immunity method,NO by Nitrate reductase method at before infarction,infarction 2h,reperfusion immediate,30min,1h,2h,4h after reperfusion.we monitored mean pulmonary artery pressure(MPAP) at the same time. The pulmonary tissue specimen fixation at the corresponding time,then pathological analysis was carried.38 rabbits was studied and the final experimental date were obtained from 27 rabbits,consisting of three groups:⑴,IR-treatment group(n=9);⑵,IR control group(n=9);⑶,sham operation group(n=9).Results:1.TNF-α,NO,ET significantly increased and NO/ET significantly decreased in IR-control group as compared with sham operation group(P﹤0.01). the value peak of TNF-α,NO,ET was showed at 30min after reperfusion. TNF-α,NO,ET significantly increased at the reperfusion stage in IR-control group as compared with the infarction stage in IR-control group(P﹤0.05);2. TNF-α,NO,ET significantly decreased and NO/ET significantly increased at the reperfusion stage in IR-treament group as compared with the reperfusion stage in IR-control group (P﹤0.01). TNF-α,NO,ET increased at the reperfusion stage in IR-treament group as compared with sham operation group (P﹤0.01). the value peak of TNF-α,NO,ET was showed at 30min after reperfusion in IR-treament group;3.MPAP significantly increased in IR-control group and IR-treatment group as compared with sham operation group at the infarction stage(P﹤0.01). MPAP did not significantly change in IR control group and IR-treatment group as compared with sham operation group at the reperfusion stage except for reperfusion immediate (P﹥0.05);4. Pulmonary tissue had significantly pathological change at the infarction stage as compared with sham operation. There was the best significantly pathological change in IR control group at 30min after reperfusion; Pulmonary tissue pathological change had significantly ameliorated in IR-treatment group as compared with IR control group at the reperfusion stage.Conclusion:1.Rabbits pulmonary embolism ischemical reperfusion injury was related with the change of TNF-α,NO,ET in the blood;2.XingDing can decrease the level of TNF-α,NO,ET and effectively relieve pulmonary ischemical reperfusion injury;3.Further definite the key point of MPAP change after pulmonary embolism is determined by pneumoangiogram bed mechanical obstruction factor;4. The release of active materials due to ischemical reperfusion can further produce the pathological harm in pulmonary tissue.
