Objective: To evaluate the effects of Ginkgo biloba Extract (EGb761) on theproliferation and the neuron differentiation of Human Adipose-Derived Stem Cells(hADSCs).Methods: Using the CCK-8assay for the detection of different concentration ofEGb761(40mg/L,80mg/L,120mg/L) on the hADSCs proliferation; containingdifferent concentration of EGb761(40mg/L,80mg/L,120mg/L) combined neuralinducing factor (20ng/ml bFGF,20ng/ml EGF,20ng/ml BDNF) on hADSCs toinduce the neuron differentiation. Observation of the morphological changes;Immunofluorescence detection of Nestin, NSE, GFAP to3D,7d,10d,14d expression,counting cells positive rate.Results: EGb761on the proliferation of hADSCs has the time difference,1d hasno significant difference,3d has remarkable promotion,5d has romote flattenedproliferation, cells proliferation rate reduce on7d; containing various concentrationEGb761induced by a combination of factors to promote hADSCs morphological neuraldifferentiation,compared with the positive control group at each time point ofpromoting the expression of Nestin, and the concentration dependence of the trend;promoting NSE expression, no obvious concentration dependence;the expression ofGFAP positive control with no difference.Conclusion EGb761on the hADSCs proliferation has promoting effect,especially on3d; EGb761significantly promoted hADSCs neural differentiation, do notpromote to glial cell differentiation. |
