Objective To investigate the protective effects and the mechanisms of the ZIWEI granule on sodium hydroxide-induced gastric mucosal lesion in SD rats.Methods Dividing sixty healthy SD rats(male 50%, female 50%) into six groups in random: the normal group; the injured group; the smecta group; the low dose group; the middle dose group and the high dose group. The smecta group, the low does group, the middle does group and the high does group were pretreated with smecta and ziwei granule separately and 3 hours later poured with 0.2N sodium hydroxide to lead to acute gastric mucosa lesion. After 45 minutes, we can measure the injured index of gastric mucosa, the concentration of EGF and PGE2 in both blood and gastric mucosa, and we can observe the pathological changes under microscope to evaluate the extent of injury and compare the effects of drugs.Result The datas of the experiments arranged successively as follows, the injured index of gastric mucosa: 0, 48.80 5.98, 18.30 9.96, 18.50 2.17, 7.17 4.07, 4.10 2.08 respectively; The concentration of epidermal growth factor in blood: 0.53 0.057, 0.38 0.092, 0.45 0.100, 0.47 0.11, 0.47 0.11, 0.52 0.14 (ng/ml) respectively; The concentration of prostaglandin in blood: 178.40 0.82, 118.54 6.94, 178.51 28.87, 173.84 25.03, 212.03 31.19, 220.29 32.42 (pg/ml) respectively; The concentration of epidermal growth factor in gastric mucosal: 2.71 0.46,1.73 0.37, 2.66 0.84, 2.31 0.62, 2.56 0.32, 3.19 0.56 (ng/ml) respectively; The concentration of prostaglandin in gastric mucosal: 217.36 9.66, 147.87 14.75, 194.78 23.29, 196.02 36.21, 230.99 13.76, 235.18 43.90 (pg/ml) respectively. A remarkable protection was founded in gastric mucosa pretreated with ZIWEI granule. Compared with the injured group, the injured index of gastric mucosa descend significantly (p<0.001), the concentration of prostaglandin in blood increased greatly (p<0.05), the concentration of epidermal growth factor in gastric mucosal increased greatly(p<0.05), the concentration of prostaglandin in gastric mucosal increased significantly (p<0.01). The concentration of epidermal growth factor in blood of all pretreative group is higher than the injured group, but the high does group and the injured group exist great difference(p<0.05). And the histological sections also revealed that the degree of inflammation and injury of the low does group; the middle does group and the high does group obviously reduced.Conclusion The gastric mucosal protection of ZIWEI granule against sodium hydroxide-induced injury in rats is significant. The effects are implemented by increasing the concentration of epidermal growth factor and prostaglandin in both blood and gastric mucosal.
