Objective: TO study expressions and correlations of p53,bcl-2,VEGF,MVD in NSCLC and discuss their significance in carcinogenesis and progression of lung cancer, as well as their relations with lymph node metastasis,angiogenesis and prognosis. Methods: 56 surgically resected patients with NSCLC combinated with radiotherapy±chemotherapy,between 1996 and 2003,were enrollde in the study group.The expressions of p53,bcl-2,VEGF and MVD were tested by immunohistochemical SP method. The results were reciprocally analyzed and compared with lymph node metastasis,angiogenesis and prognosis.Result: p53,bcl-2,and VEGF positive stained were a greater or lesser degree detected in all of the 56 cases.Positive expressions of p53,bcl-2,VEGF were found 55.36%(31/56),53.57%(30/56),57.17%(32/56) respectively. The mean MVD of all of the 56 cases was 6~69(25.80±10.55)/×400.There was no statistic difference between their expressions and age,sex,clinical stage,pathological type,pathological differentiation(P>0.05).But there was significant positive correlation between MVD and lymph node metastasis(P<0.001).Analyzed on the basis of 40 follow-up cases, there was no statistic difference of the expressions of p53,bcl-2,VEGF,MVD between survival time <1 year group and ≥1 year group(P>0.05). The expression of bcl-2 had no correlation with p53,VEGF,MVD in NSCLC.But p53,VEGF,MVD had significant positive correlation reciprocally(p<0.001).MVD in p53 positive stained group was 31.03±10.27,in another group 19.32±6.64,there was significant difference (P<0.001).MVD in VEGF positive and negative group was 31.38±9.60,18.38±6.41 respectively,as well as significant difference (P<0.001).There was no correlation between bcl-2 and p53,VEGF,MVD(P>0.05).Conclusions: p53,bcl-2,VEGF and MVD had relationship with carcinogenesis and progression of NSCLC. p53,VEGF and MVD had significant positive correlation,it was made clear that p53,VEGF synergistic participated in the regulation of neoangiogenesis process. Significant positive correlation between MVD and lymph node metastasis indicated that there was probably relation between neoangiogenesis and lymph node metastasis,to some extent. The expression of bcl-2 had no association with p53,MVD,VEGF ,we can suppose that bcl-2 had control mechanism of apoptosis which is different from p53. There was no statistic difference between their expressions of p53,bcl-2,VEGF,MVD and age,sex,clinical Stage,pathological type,pathological differentiation,1 year survival rate(P>0.05),but it was necessary to study the correlation between the four tumor markers and long-term survival further.
