[objective] The molecule mechanisms of brain edema after intracerebral hemorrhage(ICH) is complicated,And aquaporin(AQP) is the main clue to investigate the molecule reason. We produced the experimental intracerebral hemorrhage model in rats by infusing bacterial collagenase â…£into the right caudate nucleus, characterized the lesion by neurological severity score, brain water content(BWC), behavior and AQP9 expression. In intervention group, pyrrolidine dithiocarbamate was injected into peritoneum to observe the change of brain water content and AQP9 expression, to investigate the relationship between AQP9 and brain edema around the hematoma.[methods] The healthy Sprague-Dawley (weight 200-350g) rats were individed into normal group, control group, ICH group and intervention group. In ICH group and intervention group the intracerebral hemorrhage model is established by a microinfusion pump comtaining 0.065U collagenase (Type â…£ ) infused into caudate nucleus. And collagenase is not injected in control group and in normal group. For the intervention group, pyrrolidine dithiocarbamate was injected into peritoneum (200mg/100g, ip, daily) after 2 hour of ICH, and injected for 1 day, 2 days, 3 days and 5 days according to the study. We observe the neurological deficts, harvest the brain samples and examine the bain water content and expression of AQP9 by immunohistochemistry at the different time. [result] 1.AQP9 labeling is observed on astrocytes of glia limitans and aroud the ventricles, also resides on astrocytes of ventricles choroids plexis, in the hippocampus, in various hypothalamic nuclei in normal and control group. 2. The neurological severity score, brain water content and AQP9 expression in the control group is not differen from the normal group. 3. After ICH, the expression of AQP9 and the brain water content in hematoma side of ICH reinforce, beginning at the 4th hour. At the 72th hour, AQP9 is strongest expressed and the brain water content is highest. The brain water content and the expression of AQP9 in hematoma side is higher than the opposite side. 4. The expression of AQP9 and the brain water content are reduced after the intervention of pyrrolidine dithiocarbamate.[conclusions] The brain water content and the expression of AQP9 begin to rise at the same time after ICH, and fall parallelly after using the intervention. AQP9 is included in the brain edema after ICH.
