| Objective: To separate and purify the flavonoids andterpenoids of Yulangsan (YLS);And to screen the chief effectivecomponents of antihypertensive by studying their pharmacodynamicson cardiovascular system in rats. Methods : (1) The flavonoids andterpenoid were isolated from the roots of YLS with 60~70% ethanol andpurified by AB-8 macroporous resin. UV,HPLC,IR were used for thestructural identification. (2) The flavonoid,terpenoid and polysaccharide( the polysaccharide was isolated by other members ) were administratedby injection into vein(i.v.) to monitor the changes of systolic bloodpressure(SP),diastolic blood pressure(DP),and mean arterial pressure(MP) by cannulation in common carotid artery. Then 70 spontaneouslyhypertensive rats (SHRs) were divided into 7 groups: nifedipin group,three mixed -compound (MC,it was extracted from the roots of YLS byhot water . ) groups,flavonoid group,terpenoid group and saline group.And ALC-NIBP biological signal analysis system were used to measuredtail blood pressure of the waking SHRs every other 10 days. (3)MPA-Cardiac Function Acquistion and Analysi System(MPA-CFS) wereused to observe and record HR,LVSP,LVEDP,dp/dtmax,- dp/dtmaxin anesthetized Wistar rats before and after the administration respectively.Results: (1) The extractive flavonoid showed as yellow needle stylecrystal. It was easily dissolved in methanol, ethanol and chloroform etc.The Rf value of ployamide thin layer chromatography was 0.59. Theresult of high performance liquid chromatography (HPLC) showed asingle peak whatever the mobil phase was methanol-water-phosphateacid(55:44.5:0.5) or methanol-water(70:30) or acetonitrile-water(60: 540). (2) The terpeniod that was isolated by AB-8 macroporous resin waslight yellow. It was easily dissolved in water and hot methanol. The resultof Lieberman-Burchard reaction indicated it was triterpeniod. AndUV-spectrum analyses showed that there was a maximal ultravioletabsorption peak at 218nm. (3) The flavonoid and terpeniod couldsignificantly decrease the carotid blood pressure in Wistar rats and SHRs.(4) In chronic antihypertensive experiment, The oral administration of theflavonoid or terpeniod or MC for 20~30 days could significantlydecrease the tail blood pressure of SHRs. The maximum reduction inblood pressure induced by the flavonoid or terpeniod or MC was a littlelower than that by nifedipin. But the duration of BP-reducing effect ofeach of them was better than that of nifedipin. (5) The flavonoid andterpeniod could significantly decrease LVSP,dp/dtmax,ET,At(CFL),As(CFL),Ad(CFL), lengthen IT,STI and reduce the amplitude ofcardiac contractility. Conclusion: The flavonoid and terpeniod were themain chemical compositions of YLS, And the inhibition of myocardialcontractility and decrease in HR were the main causes resulting in thedecrease of the blood pressure. |
