The uterine cervical carcinomal is one of the most common malignant tumors in women, the mortality of the disease has been keeping highly. Mestatasis and the trend is become more and more younger. Mestatasis and transition is the main cause of carcinomal patientscieath . Recently, the evidence has demonstrated that the over-expressions of matrix metalloproteinase make a new road for tumor cells invading and transiting.Matrix metalloproteinases are a familly of Zine dependent metalloprotemas-es which participate in the degradation of collagen and other extracellular matrix macromolecules. There are twenty-one kind of proteinases , collagen enzymes IV, designated MMP-2, MMP-9 are studied much more popularly . MMP-2, MMP-9 exist commonly in the urinary, gastric, breaseand liver, they are linked closely with the stages of cases. The enzymes actives of MMP-2, MMP-9 can be inhibited specially by actors ( TIMPs). TTMPs is the nature inhibitor living in peoples' bodies, they are bound with MMPs by complex and keep a activing balance usually to express important physical function.This study population consisted of 33 patients , diagnosed with squanous cell carcinoma of the uterine cervix , we took a kind of technique called RT-PCR for the rapid and sensitive detection of mRNAs in tissue . The expression of MMP-2, MMP-9 and TIMP-1, TIMP-2 in cervical carcinomal tissues are determined and MMP/TIMP ratios are compared in the cervical cancers with clinical stage and histry grade. To study its function in cervical carcinomal.Materials and Methods1 Materials We had analysed 33 cases of uterine cervical squamous cell carcinomas , 10 cases of normal cervical tissues at contral for the presence of MMP-2 MMP-9 , TIMP-2 TIMP-1 using RT-PCR . The results were analysed for the possible correlation with diseases outcome .2 Reagents . Trizol and reverse transcription kit were purchased from TaKa-Ra Inc . The primers of MMPs TIMPs and action mRNA were synthesized by Beijing AoKe biological Inc.RT-PCR and DNA ladder reagents were proviced by the congenital malformation lab of the second clinical college of China Medical University .3 Methods Semiquatitative reverse transcriptase polymerase chain reaction technique .Statistical analysis was performed using SPSS for windows 10. 0 software . The data of the experiment was expressed as means . And comparisons of means were carried out by t test .Result1 Expression level of MMP-2 MMP-9 in normal cervical tissue and cervical carcinomal tissue were as follow : 76. 3 15.9 , 96. 2 4. 6 and 103.1 29.9 , 112. 5 20. 3 . The expression level in cervical cancers of MMP-2 MMP-9 were significantly higher than that in normal cervical tissue (P <0.01) .2 Expression level of TIMP-1, TIMP-2 in nomal cervical tissue and cervical carcinomal tissue were as follow: 106. 5 8. 3, 110. 3 7. 3 and 15. 9 4. 8, 39.8 27. 9. The expression level of TIMP-1 TIMP-2 were significantly lower than that in normal cervical tissue ( P <0.01).3 MMP-2/TIMP-2 and MMP-9/TIMP-1 ratios were becoming higher with cervical carcinomal advance.DiscussionThe dissemination of malignant neoplasms is a multistep process , involving degradation of subepithelial basement membranes and the extracellular matrix ( ECM) . Matrix metalloproteinases ( MMPs) , a family of zine-dependent enzymes , appear to play a key role in turner invasion and metastasis, being able to degrade most ECM components.The role of MMP-2 MMP-9 and tissue inhibitor of TIMP-2 TIMP-1 in the invasive and metastatic processes, as well as their functional interrelating effects , are the subject of a growing number of studies. In 1996, Grignon DJ, et al studied that breaking the balance of MMPs and TIMPs in tumor is a advanced factor of turner. In 2002, Jiang YangFu Gelderg studied that lower expression of TIMP-1 TIMP-2 is associated with metastasis.
