Objiective To investigate the effect of Cordycepssinensis on the renal ECMof Wistar rats with progressive glomerularsclerosis in rats and probe into its machenics. Methods 36 Healthy 50-days-old Wistar rats were equally divided into three groups by chance:normal control group, progressive glomerularsclesis group made by 5/6 nephrectomy and Cordycepssinensis-treated progressive glomerularsclerosis group wich would be treated by Cordycepssinensister 5/6 nephrectomy. 4 weeks and 9 weeks after 5/6 kidneys being nephrectomized in the progressive glomerularsclesis group and after sham operations in the control group, half rats of each group were killed, renal pathological changes were evaluated and immunohistochemistry was used to examine the expression of fibronectin(FN), collagen IV, matrix metalloproteinase (MMP-2), tissue inhibitor of metalloproteinase -l(TIMP-l) and TIMP-2. RT-PCR was used to examine the MMP-2.TIMP-1 and TIMP-2 mRNA in the kidneys. Results (1) 4 weeks after operation urinary albumin excretion and TIMP-2 of Cordycepssinensis-treated progressive glomerularsclerosis rats was substantially deseased(P<0.05), Compared with the untreated 5/6 nephrectomized group. (2) 9 weeks after operation , nrinary albumin excretion(P<0.05), serum cholesterin(P<0.05) and low density lipoprotein(P<0.05) of Cordycepssinensis-treated progressive glomerularsclerosis group were decreased, compared with the untreated 5/6 nephrectomiz ed group. Pathohistological study revealed that, there were less glomerular matrix, type IV and FN in Cordycepssinensis-treated progressive glomerularsclerosis group than progressive glomerularsclesis group.The levels of TIMP-lmRNA(P<0. 05),TIMP-1(P<0. 01), TIMP-2mRNA(P<0. 05) and TIMP-2 were remarkly reduced in Cordycepssinensis-treated progressive glomerularsclerosis group. Conclusion Cordycepssinensis can reduce proteinuria , serum cholesterin, low density lipoprotein and adjust, the balance of MMPs and TIMPs in 5/6 nephrectomized rats. Cordycepssinensis significantly alleviates theaccumulation of extracellular matrix and prevents renal injury following 5/6 nephrectomy.
