Objective To study the significance of matrix metalloproteinase-2 (MMP-2) and tissue inhibitor of metalloproteinase-2 (TIMP-2) expressing in craniopharyngiomas.Methods Collecting the clinic data of 49 craniopharyngiomas, including gender, age, tumor type, disease recurrence, and scaling the MRI by Qi's scale before operation. Using SABC immunohistochemistry to detect the expression of MMP-2 and TIMP-2 in 49 craniopharyngiomas. Formalin-fixed paraffin-embedded tissue sections from 49 craniopharyngiomas were immunostained by a manual method using monoclonal antibodies against MMP-2 and TIMP-2. Immunoreactivity was semiquantitatively scored based on stain intensity and positive cell, and results were analysised with gender, ages, pathologic type, and disease recurrence by SPSS 10.0.Results MMP- 2 positive is localized in the cytoplasm of tumor cells, matrix closing tumor expressed negative. Both adamantinomatous and papillary expressed MMP-2. 28 cases of 49 craniopharyngiomas had positive expression (57.1%) and 21 cases negative (42.9%) (P>0.05). TIMP-2 positive is localized in the stroma cell near the tumor, but tumor cells didn't express. Both adamantinomatous and papillary poorly expressed MMP-2. 7 cases of 49 craniopharyngiomas had positive expression (14.3%) and 42 cases negative (85.7%) (<0O.05). The differenece of MMP-2/TIMP-2 expressing quantity didn't have statstics significance in gender, age, tumor pathologic type, disease recurrence and Qi's scale (P>0.05). Gender, age, tumor pathologic type, disease recurrence and Qi's scale had nonsignificant effect with MMP-2/TIMP-2 (P>0.05).Conclusion Both adamantinomatous and papillary craniopharyngiomas express MMP-2. MMP-2 can't predict invasion in craniopharyngiomas, maybe correlates with another role, including modulating tumor hormone secretion and cell proliferative. Craniopharyngioma is a low invasive tumor.Preoperative MR can help to judge the infiltrative grow of cramopharyngiomas. TIMP-2 nearly expressed in cramopharyngiomas.
