| Objective-to investigate the relationships of polymorphisms of MTHFR gene with cerebrovascular disease in young adults .Methods-this case-control study was based on 67 consecutive patients (37 males, 30 females;mean SD age 42.5 6.6 years ;range,20 years to 50 years)of young adult cerebrovascular disease.71 age-and sex- matched apparently healthy subjects (32 males,39 females;mean SD age 40.7 7.5 years ;range,20 years to 50 years) served as control.Their polymorphisms of MTHFR gene were analyzed using PCR-RFLP. Results-there are three kinds of MTHFR 677genotype: homozygous mutation (T/T), heterozygous mutation (C/T) and wild-type (C/C). The frequencies of the three genotypes were as follows: T/T, 29.85%; C/T, 47.76%; C/C, 22.39% in stroke patients and T/T, 42.25%; C/T, 47.89%; C/C, 9.86% in normal subjects, respectively. The frequency of T alleles was significantly higher in normal controls than that in stroke patients (66.20% and 53.73%, respectively) 0 There are two kinds of MTHFR 1298genotype: heterozygous mutation (A/C) and wild-type (A/A). The frequencies of the two genotypes were as follows: A/C, 43.28%; A/A, 56.72% in stroke patients and A/C, 19.72%; A/A, 80.28% among normal subjects, respectively. The frequency of C alleles was significantly higher among stroke patients than that among normal controls (21.64% and 9.86%, respectively).Conclusions-In this population, MTHFR 677 and 1298 polymorphisms are positively associated with risk of cerebrovascular disease in young adults .The frequency of 677 homozygous mutation (TT )and T alleles in MTHFR genotype is higher in Qingdao population than in others of Asia and Caucasians . This mutation maybe protect the population from stroke not hazardous factor in the most papers.But The frequency of 1298 heterozygous mutation (AC )and C alleles in MTHFR genotype is higher among stroke patients than that among normal controls, the results suggest that MTHFR 1298 polymorphisms may be a risk factor for cerebrovascular disease in young adults . |
