objective To observe the inducible expression of heat shock protein 70 (HSP70) in rat retina neurons and the eye tissue injury after intraperitoneal injection of zinc sulfate Methods Thirty-six rats were divided into 3 test groups (12rats each group), regardless sexual , by random method. The left eyes in the test groups were treated by intraperitoneal injection of zinc sulfate in different concentrations (15mmol/l, 1.5mmol/l, 0.15mmol/l),as the self antitheses groups the right eyes of all the rats were treated by intraperitoneal injection of physiological salt solution equal to the capacity of zinc sulfate. The test groups were subdivided into group 1, 2, 7and 14 days after injection. The rats were sacrificed at corresponding time and took out all the tissue in the orbits. After fixed in 10% neutral formalin, the expression of HSP70 was detected with immunohistochemical techniques. The inflammation was observed with HE staining. Results 1. Positive HSP70 Immunoreactivity was expressed in retinal ganglion cell layer and inner nucleous layer of all rats in test groups. The positive signal located to the plasma and the nuclear, especial the plasma. Low-concentration group had a more weak expression, in contrast to the middle-and high-concentration group. A few of HSP70 positive cells were found 1 day after treatment. The amount of HSP70 positive cells increased at all time. About 2 days after treatment came to its peak time and remained till 7 day, and then it declined in 14 day.2. Positive HSP70 Immunoreactivity was expressed in retinal ganglion cell layer and inner nucleous layer of all rats in the self antitheses groups, but under the amount of the test groups obviously. 3.Slight inflammatory response wasobserved in local tissue with high-concentration injection by HE staining. On the contrary, the inflammatory response in low- and middle-concentration group was untraceableAMoreover; HSP70 was inducibly expressed in cornea and optic nerve by zinc sulfate. Conclusion 1. Zinc sulfate can induce HSP70 expression in retinal neurocyte through intraperitoneal injection, as plays a vital role in pathology process, such as injury repair and tissue regeneration.2. Injection results in inflammation of high-concentration group rats, no obvious inflammation was found in low- and middle-concentration group rats.3. Middle-concentration of zinc sulfate is a proper concentration in inducing the HSP70 expression in rat retina neurons.
