Objective: Research the influence of heat shock protein-70 to defection of neural function in rat following focal cerebral ischemia and reperfusion and the expression of heat shock protein-70 after different reperfusion time.Method: Seventy-two healthy male Wistar rats were chosen and were randomly divided into four groups(18 in every group): ischemia 1 hour/reperfusion group, ischemia 3 hour/reperfusion group, sham operation group and complete ischemia group. The reperfusion time were 24 hours. The cerebral ischemia reperfusion injury models were made by the method of middle cerebral artery thread.The expression of HSP70 in peripheral blood was determined by flow cytometry and in brain tissue was determined by immunohistochemical method. The volume of cerebral infarction was determined by TTC staining method.The neural functional evaluation of the rats were on the clear-headed time after anesthesia,the time after reperfusion and the seventh day after the operation.Result: The HSP70 expression in sham operation group was very low. The TTC staining result did not show cerebral infarction.The HSP70 expression in ischemia groups were heat up obviously: The HSP70 expression in ischemia 1 hour/reperfusion group was the highest.After it was ischemia 3 hour/reperfusion group.The complete ischemia group was the lowest. It had statistical significance after the contrastive study of these groups(P<0.05). The cerebral ischemia groups'TTC staining results can see the different degrees of cerebral infarction.The defection of neural function was lower in ischemia 1 hour/reperfusion group and was the most serious in completely ischemia group.Conclusion: Ischemia and reperfusion can advance the HSP70 expression in blood and brain. If the HSP70 gets much more, the long-term neural function will recover much better.This demonstrates that HSP70 may offer protection to neural function. The HSP70 level of lymphocytes in the blood is synchronized with the HSP70 level of cerebral ischemia.
