Chemokine is a big category of cell Factors , it can Jink with the receptor of leukocyte, and activate the transfer signal of receptor, then the leukocyte starting the chemotactis action. It shows main drug value on the lymphocyte recycle, chronic inflammation reaction, allergic diseases, self-immunity diseases, resisting infection and tumour and the prevention and cure of AIDS. Up to now there are more than 50 Chemokine discovered, the molecular weight is around 8~12Kda.The molecular structure of Chemokine has high similar origin. They all include 4 conservative cysteines, forming the presence of two disulphide bonds, according to the relative position of the 4 conservative cysteines, Chemokine is separated into 2 sub-strains -a&P, they activate different sub-leukocyte groups, and have obviously selecting spec.il ization. a sub-strain (IL-8, NAP-1, NAP-2 etc) activates neutrocyte, p sub-strain (MCP-1, MCP-2, MIP-1 etc) activates monocyte and/or T-lymphocyte. Presently, most of the Chemokine studies are focus on its structure and function in-body, there are few reports on filtrating chemokine fromnatural protein outside body. Some components of snake toxin are similar with chemokine, they have the hemylosis action inside the body and cruor action outside the body. It has the same receptor with that of chemokine, both of them are G protein-coupled receptors, so it might contain some chemokine or analog in the snake toxin. We set up the chemokine filtrating system on the molecular level, then separated and purified more than 400 small peptides whose the molecular weight is similar with chemokine from more than 12 kinds of snake toxins, and made as a sample, to find out the chemokine inducing elements and new chemokine by filtrating with chemotaxis and Cytosensor. After twice filtrating the active samples by chemotaxis and Cytosensor, then eliminating the cell toxin component through the in vitro cell toxin test, the 3 samples with chemotactic capability ?knis2-2k?nis3-6?knis5-3 were obtained.
