To study the effective ingredients of traditional Chinese medicine prescription Yu ping feng, which have anti-inflammatory and immunodulatory actions, an effective part-total glucosides of Yu ping feng(TGYPF) was isolated by extraction, isolation, refinement and purification with the techniques and methods of chemistry of TCM. By use of orthogonal experimental design L9 (34) , we screen out that the best method of extraction conditions is 8 times amount of 80%EtOH, extracting solvent to reflux for 2 hour and 3 times.And the HPLC-ELSD analytical method has been developed for the assay of the Astragaloside in the main active principle Radix Astragali. The average recovery is 95.33%, with RSD of 1.52%. The method validation has also been examined. The result shows that the precision and repeatability of this method are good respectively. The method is simple, fast and accurate and can be applied to the analysis of the Astragaloside. The presence of other ingredients in the prescription was identified by TLC, respectively. It provides the scientific experiment basis for TGYPF of its quality standard.To investigate the anti-inflammatory and immunodulatory actions of total glucosides of Yu ping feng (TGYPF) and its mechanism, the acute and chronic inflammatory models were established, meanwhile a series of immumopharmacological experiments in vivo were adopted. The main results as follows: 1. Anti-inflammatory actions of TGYPFTGYPF (24, 48 , 96 mg-kg-1, ig ) could significantly inhibit the mice ear swelling induced by dimethylbenzene, so could TGYPF (16, 32, 64 mg-kg-1, ig ) inhibit the rat palm swelling and the rat cotton pellet granuloma induced by carregeenin, andcotton pellet wool respectively. The results showed TSYPF have a remarkableanti-inflammatory action.2. Immunoregulatory actions of TGYPFTGYPF (24 , 48 , 96mg-kg-1, ig) apparently improved the hemolysin level in serum and splencyte in mice immunosuppressed by Cy. Delayed-type hypersensitivity (DTH), which suppressed by Cy, was enhanced by TGYPF (24 , 48 , 96 mg-kg-1, ig) in mice. These results suggested TGYPF could recover nonspecific, humoral and cellular immune function in mice with immunosuppression. TGYPF (48 , 96 mg-kg-1, ig) significantly increased both CD4+ and CD8+ cell, especially influenced on CD4+ cell, which rendered the ascending the ratio of CD4+/ CD8+ cell. The results suggested the immunoregulatory actions of TGYPF might be brought about via regulating T cell subtype.
