In this paper,the extraction and purification process of Astragalus saponin was optimized to establish a high efficient and stable saponin extraction and purification process,and the study on the effect of saponin-like components in vitro antiviral?Coxsackie B5?was carried out.The contents of the study include: 1.Through the choice of extracting solvents from Radix astragali,at the same time,the determination method of the content of total saponins and Astragaloside IV.In Astragalus Membranaceus was established,the method was stable and reliable,and the content of total saponins and Astragaloside IV in Astragalus Membranaceus was determined.Finally,the extraction solvent of this subject was selected as water.2.By means of orthogonal test,to determine the extraction process of radix astragali for boiling 2times,the first time add 9 times times the amount of water,fry 2 hours,the second add 7 times times the amount of water,fry for 1 hours;purification process using potassium dihydrogen phosphate-anhydrous ethanol aqueous phase system,the total saponins in Astragalus were purified;Finally,polyamide column was used to separate the total saponins from Radix Astragali,and after polyamide was activated,2.5g original drug?wet method?was activated in 100 ml,and 1h,with 3BV water,25%ethanol solution,50% Ethanol Solution elution,elution speed is 1 0BV.Results after extracting and purifying the medicinal herbs,Astragaloside IV was the main reference index,the total transfer rate of astragaloside 77.1% was 73.3%,in which the transfer rate of Astragaloside IV.Was 83.77%,the transfer rate of Astragaloside was 84.32%,and the purity of total saponin was 46.02% after purified by double aqueous phase.The transfer rate of Astragaloside 92.05% was 87% in total saponins of Astragalus membranaceus after polyamide column.The purity of total saponins was 87.2% after purification and separation of polyamide.3.By introducing the "one-test-multi-evaluation" method,the establishment of the quality standard for the effective parts of Astragaloside IV.By using astragaloside internal,and calculating the variable coefficients of each component,the comparison between the relative retention value and the result data and the external standard method,the results show that the HPLC method is simple and reliable,and can be used to determine the content of Astragalus saponin Shan?,Astragaloside Shan?,Astragaloside Shan?,Astragaloside Shan ??astragaloside?and saponins Shan? and Astragalus saponin Shan?.The determination of saponin components in radix astragali by "one-test-Multiple evaluation" method showed that rsd of relative retention value was high,but there was no difference between the calculated content data and the foreign standard method.According to the Chinese Pharmacopoeia of a 2015 edition of Astragalus membranaceus astragaloside requirements,the requirements of radix Astragali according to dry product calculation,containing astragaloside?C41H68O14?should not be less than 0.04%.Therefore contain Astragalus saponin Shan not less than 0.031%,Astragalus saponin Shan not less than 0.032%,Astragalus saponin Shan not less than 0.014%,Astragalus saponin Shan not less than0.022%,Astragalus saponin Shan not less than 0.024%.4.To explore the effect of the original medicine,the total saponins of Astragalus?50% alcohol elution?and the composition of polyamide water on the immune function of mice.Results The Astragalus membranaceus could significantly improve the immune ability of mice,and could promote the growth of immune organs and the content of cytokines in mice.The results showed that Astragalus saponin could promote the immune ability of mice.5.The original medicine,the total saponins of Astragalus?50% alcohol elution Group?,in vitro antiviral study of polyamide washed off components showed that the antiviral effect of the central medicine and the total saponins of Astragalus membranaceus was obvious in the antiviral experiment in vitro,the protective effectof the cell and the effect of antiviral adsorption in four experiments were better than that of cell proliferation and direct antiviral action.The experimental results can confirm the effect of Astragalus saponin on anti-Coxsackie?B5?virus in vitro.
